Description of key information

The objective of the study was to evaluate Pigment Orange 5 for its ability to generate toxic effects in Raphidocelis subcapitata during an exposure period of 72 hours and, if possible, to determine the NOEC, EC10, EC20 and EC50 for both inhibition of growth rate and inhibition of yield. A combined limit/range-finding test was performed and the following nominal test concentrations were used: 0.063, 0.63 and 6.3 μg/L. Under the conditions of the present study with Raphidocelis subcapitata, no inhibition of growth rate or inhibition of yield was recorded at any of the loading rates of Pigment Orange 5 tested. The 72h-EC50 for growth rate inhibition (ERC50) and yield inhibition (EYC50) exceeded a loading rate of 6.3 μg/L. The 72h-NOEC for both growth rate inhibition and yield inhibition was 6.3 μg/L. Due to the very low solubility of Pigment Orange 5 in water, concentration levels that might be toxic for algae could not be reached. The study met the validity criteria prescribed by the study plan and was considered valid.

Key value for chemical safety assessment

EC50 for freshwater algae:
6.3 µg/L
EC10 or NOEC for freshwater algae:
