Key value for chemical safety assessment

Skin sensitisation

Endpoint conclusion
Additional information:

In the chosen key study for skin sensitization, a guinea pig maximization test according to OECD TG 406 and GLP, Himalayan White spotted guinea pigs have been treated with retinyl propionate intradermally (5% in olive oil) and topically (undiluted) for induction (DSM_B154702). Challenge exposure was performed topically for 24 hours using 30% retinyl propionate in olive oil and reactions were observed immediately, 24 and 48 hours post application.

No positive reactions were evident after the challenge in vehicle controls. In the test substance treated group, dermal reactions after challenge application were observed in 1/19 animals. One female of the test substance treated group died spontaneously on day 21 of the test.

Based on the data reported, it can be concluded that retinyl propionate shows no dermal sensitization under the test conditions chosen.

Justification for classification or non-classification

The present data on dermal sensitization do not fulfill the criteria laid down in 67/548/EEC and 1272/2008/EEC, and therefore, a non-classification is warranted.
