Please be aware that this old REACH registration data factsheet is no longer maintained; it remains frozen as of 19th May 2023.

The new ECHA CHEM database has been released by ECHA, and it now contains all REACH registration data. There are more details on the transition of ECHA's published data to ECHA CHEM here.

Description of key information

Basic toxicokinetics

ZDDPs are expected to have low oral and dermal absorption based on modeling, toxicity data, and physiochemical properties. Upon ingestion, ZDDPs are expected to be transformed in the GI fluid to the corresponding alkyl dithiophosphate ester (dissociated from the Zn). Any alkyl dithiophosphate ester that is bioavailable will undergo metabolism consistently across the category, ultimately being metabolized to the corresponding starting alkyl alcohol followed by typical alcohol metabolism.

Stomach simulation study

A critical question about the ZDDP chemical category is whether the amount of basic vs. neutral ZDDP will influence the toxicity and, hence, whether this is a variable that requires consideration in e.g., the boundary composition for the category. These results demonstrate that, upon ingestion, ZDDPs will rapidly and completely convert from the basic form to the neutral form. This information is critical to developing the Intelligent Testing Strategy for the ZDDP category because it demonstrates that the basic form of ZDDPs are not relevant from a toxicity perspective. Specifically, upon ingestion there is transformation of all ZDDPs to the neutral form which becomes the common denominator.
