Study period:
2009-Nov-6
Reliability:
2 (reliable with restrictions)
Rationale for reliability incl. deficiencies:
guideline study with acceptable restrictions
Qualifier:
according to guideline
Guideline:
OECD Guideline 203 (Fish, Acute Toxicity Test)
Qualifier:
according to guideline
Guideline:
EPA OPPTS 850.1075 (Freshwater and Saltwater Fish Acute Toxicity Test)
GLP compliance:
no
Analytical monitoring:
yes
Vehicle:
yes
Test organisms (species):
Danio rerio (previous name: Brachydanio rerio)
Details on test organisms:
Zebrafish (Brachydanio rerio) were purchased from a local fish farm. All fishes were acclimated for at least 12 days in a plastic tank (60 cm X 38 cm X 38 cm). Reconstituted water with aeration was used during the acclimation and throughout the study. The dissolved oxygen of the water was maintained above 80% of air saturation with aeration. Water temperature was maintained at 22±2°C, and the photoperiod was set as 12-hour light and 12-hour dark. All fishes were fed daily with commercial fish feed. Feeding was stopped at about 18-24 hours before the study.
Test type:
static
Water media type:
freshwater
Total exposure duration:
96 h
Test temperature:
The temperature of the test solution ranged from 21.2°C to 21.5°C.
pH:
The pH of the test solution ranged from 7.28 to 7.67 and the control solution range from 7.20 to 7.51, respectively.
Dissolved oxygen:
The dissolved oxygen of the test solution ranged from 7.4 mg/L to 7.5 mg/L, whereas the control solution ranged from 7.3 mg/L to 7.7 mg/L.
Details on test conditions:
48 hr before experiment, the glass aquarium (30 cm X 18 cm X 20 cm) was filled with 7 L reconstituted water. The dissolved oxygen of the water was maintained with continuing aeration.
About 18-24 hr before experiment, 10 fishes were transferred from the acclimation plastic tank to the test aquarium. There were 10 fishes in each aquarium.
The test solution was prepared freshly by dissolved 700 mg of test article in 7 L of reconstituted water. Another group of 10 fishes were exposed to the reconstituted water without test article for serving as blank control.
Mortality of the fishes was recorded at 3, 6, 24, 48, 72 and 96 hours after beginning of the study.
The measurement of the pH, temperature, and dissolved oxygen were carried out at the beginning of the test and 24, 48, 72 and 96 hours after beginning of the study. A mobel InoLab 720 pH meter (WTW, Weilheim, Germany) and a model SM600 dissolved oxygen meter (Milwaukee, Gallarate, Italy) were used to determine the pH and dissolved oxygen.
At the end of the study, head to tail-base length and body weight of test and control group fishes were determined.
Duration:
96 h
Dose descriptor:
LC50
Effect conc.:
> 100 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Basis for effect:
mortality (fish)
Details on results:
The average head to tail-base length of the test group was 2.36±0.18 cm, where the control group was 2.36±0.18 cm. The average body weight of the test group was 0.30±0.05 g, whereas the control group was 0.26±0.04 g.
Sublethal observations / clinical signs:

All of the 10 fishes in test group and control group survived after exposure to test or control solution for 96 hours.

Validity criteria fulfilled:
not specified
Conclusions:
All of the control and test group fish were survived at the end of the study. The findings indicated that the 96-hour LC50 of the test substance is greater than 100 mg/L under the condition designed of this study.
Executive summary:

The study was conducted in accordance with OECD 203 to determine the 96-hour LC50 (median lethal concentration) of the test substance (Batch no. R6108507). A limit test was conducted at the concentration of 100 mg/L with zebrafish (Brachydanio rerio). The test solution was prepared by dissolved 700 mg of test article in 7 L of reconstituted water. A group of 10 fishes were exposed to the test solution for 96 hours. Another group of 10 fishes were exposed to reconstituted water and served as control group. The mortality of fish was recorded at 3, 6, 24, 48, 72 and 96 hours. The results indicated that all of the control and test group fish were survived at the end of the study. The above findings indicated that the 96-hour LC50 of the test substance is greater than 100 mg/L under the conditions of the study.
