An acute toxicity study inDaphnia magnawith the read across substance (2-Propenoic acid, 2-methyl-, 2-hydroxyethyl ester, reaction products with phosphorus oxide (P2O5)) was conducted according to OECD Guideline 202, EU Method C.2 and ISO International Standard 6341 (1996), in compliance with GLP. Based on the findings of a range finding test, twenty Daphnids per concentration (four replicates, 5 per vessel) were exposed to a blank control and a test substance concentration of 100 mg/L for 48 h, applying a semi-static design. Test substance concentrations were verified analytically with HPLC-UV at t= 0 h and t= 24 h from freshly prepared solutions and and at t= 24 h and t= 48 h from 24 h-old solutions. Only 5% immobility was observed at 100 mg/L. This was not considered treatment related as a maximum response of 10% is acceptable for the control. Analytical measurements indicated recoveries ranging between 93 and 97% of nominal concentrations. All the validity criteria was met. Hence, the EC50 of the substance was determined to be >100 mg/L (Migchielsen IMJ, 2004).
