In Article 13 of Regulation (EC) No 1907/2006, it is laid down that information on intrinsic properties of substances may be generated by means other than tests, provided that the conditions set out in Annex XI (of the same Regulation) are met.

 According to Annex XI of Regulation (EC) No 1907/2006 (Q)SAR results can be used if (1) the scientific validity of the (Q)SAR model has been established, (2) the substance falls within the applicability domain of the (Q)SAR model, (3) the results are adequate for the purpose of classification and labeling and/or risk assessment and (4) adequate and reliable documentation of the applied method is provided.

 For the assessment of the reaction mass of 4-methoxyphenol and 4-methoxy-cyclohexanone and 4-methoxy-cyclohexanol (EC 947-389-1; Mochon roh) (Q)SAR results were used to assess the acute toxicity to aquatic invertebrates. The criteria listed in Annex XI of Regulation (EC) No 1907/2006 are considered to be adequately fulfilled and therefore the endpoint(s) sufficiently covered and suitable for risk assessment.

Assessment:

No experimental data on the short-term toxicity to fish of Mochon roh are available. In order to assess the short-term toxicity potential of the test substance, the read-across data for three representative constituents of the substance.

The 48-h acute toxicity of CAS 150 -76 -5 to Daphnia magna was studied under static conditions. The GLP study was performed according to OECD 202 guideline. The 48- hour EC50 was 3.00 mg/L (measured initial, ECHA, desseminated data, 2018).

Based on the results of this study, the substance is considered to be acutely toxic to invertebrates.

A GLP OECD guideline 202 study was performed with CAS 13482-23-0. According to this static study a 48-h EC50 value was determined to be higher than 100 mg/L. (measured, ECHA, desseminated data). Therefore, the substance is considered to be acutely not harmful to aquatic invertebrates.

There are no experimental data for the third constituent 4 -methoxycyclohexan-1 -ol (CAS 18068-06-9); therefore, the toxicity of the substance to aquatic invertebrates was estimated with QSAR models: US EPA T.E.S.T v4.2.1, ECOSAR v1.11 and the QSAR Toolbox v4.2. The substance was within the applicability domain of these models. According to T.E.S.T. the 48h EC50 value was 211 mg/L. The 48 -h EC50 value was estimated by ECOSAR at 652 mg/L. A trend analysis on data from the QSAR Toolbox yielded an EC50 of 2590 mg/L. The constituent is considered to be with high probability acutely not harmful to aquatic invertebrates.

The reaction mass of 4-methoxyphenol and 4-methoxy-cyclohexanone and 4-methoxy-cyclohexanol (EC 947-389-1; Mochon roh) is a mixture of 4-methoxycyclohexanone (CAS 13482-23-0), 4-Methoxycyclohexan-1-ol (CAS 18068-06-9), Mequinol (CAS 150-76-5). The assessment follows a read-across approach based on the experimental and calculated data for its constituents. It can be concluded that Mochon, roh is acutely toxic to aquatic invertebrates.
