PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method: The test substance was stirred in demineralized water for about 20 h at approximately 20 ± 2 °C. Undissolved test substance was removed by centrifugation. The eluate had a tinge of green. The nominal concentration of the eluate was 125 (mg/l).

For growth rate and biomass, the EC50 after 72h lies above the water solubility limit.  Due to the test design the raw eluate has to be diluted (8:1:1) due to the addition of algal suspension and enriched medium.

At the highest technically feasible concentration of the eluate (due to the test design, 80% of the water solubility limit) 9.1 % effect on growth rate and 28.4% effect on biomass appeared. The NOEC lies at 1/10 of the water solubility limit for both endpoints

The substance is not toxic to aquatic algae, with an ErC50 > 100 mg/L and an ErC10 > 100 mg/L.

Additional information

All substances in this category share a similar structure with a molecular weight > 500 g/mol. Based on the high molecular weight and insolubility, the substances in this category will not be able to penetrate biological membranes. Thus, no systemic exposure is expected. This is confirmed by the environmental studies on aquatic algae for one substance in this category: CAS 1328-53-6. The environmental toxicity data can be used for read-across to all substances in this category.


The study on green algae Desmodesmus subspicatusas was performed according to OECD TG 201. The test concentrations were not analytically verified. The 72-h ErC50 and ErC10 were determined to be > 100 mg/L (the highest concentration tested) (BASF 2000, K2).


The determined ErC50 and ErC10 values were significantly higher than the water solubility of the test substance. Based on the available information, it is concluded that all substances in this category are not toxic to aquatic algae with an ErC50 > 100 mg/L and an ErC10 > 100 mg/L.
