Short-term toxicity to fish

Description of key information

The short-term toxicity to fish was examined in a study according to OECD TG 203, and in compliance with GLP (Huntingdon, 1986). In this study, groups of 10 rainbow trout were exposed to nominal concentrations of 0 (control), 10, 18, 32, 56, and 100 mg/L mg/L for 96 hours under semi-static conditions (24-h renewal of test media). A stock solution (1:1) of test substance in 30% Tween 80 (non-ionic surfactant and emulsifier) / dimethylformamide was prepared and the range of test solutions prepared by direct dilution of this stock. Test substance analysis was performed in fresh medium at the start of the test (fresh medium), just before the first medium renewal at 24 hours, and after 96 hours (old medium). As measured concentrations in the 24 -hour old test medium were found to be not within ± 20% of nominal (ca. 50 - 70% recovery), geometric mean measured concentrations were calculated. Geomean measured concentrations were 0 (control), 7.68, 13.76, 21.14, 42.39, and 82.28 mg/L. Fish mortality was recorded throughout the test and the cumulative mortality at 96 hours determined. The validity criteria of the test were met. At the lowest test concentration of 7.68 mg/L no mortality occurred. After 96 hours exposure, at test substance concentrations of 13.76 and 24.14 mg/L, mortality in fish was 20% and 40%, respectively. At the two highest test concentrations, 100% mortality was recorded. Based on these findings the 96 -h LC50 value was determined at 22 mg/L (95% C.L. 17.35 - 29.05 mg/L), based on geometric mean measured concentrations.
