Validity

There were no immobilized animal in the control group and the dissolved oxygen concentration at the end of the test in control and test vessels was more than 3 mg/L. All validity criteria were within acceptable limits and therefore the study can be considered as valid.

Concentrations of the test item

Concentration of Reactive Red F08-0146 was analytically determined at the start and at the end of each renewal period.

Measured concentrations were 106.6 and 110.6 mg/L at the start and 61.5 and 79.4 mg/L at the end of renewal periods. Concentrations of test item deviated more 20 per cent from the initial values at the end of the renewal periods, however geometric mean value of measured concentrations (87.1 mg/L) was within ±20 per cent of the nominal concentration. Biological results are reported separately based on nominal as well as measured value.

Immobilisation

The number of immobilised animals and the percentage of immobility were determined at the 24thand 48thhour. There was no immobilisation in 20 daphnids exposed to each group. In addition to immobility, no abnormal behaviour or appearance of test animals was detected.

Acute toxicity of Reactive Red F08-0146 was assessed with Acute Immobilisation Test on Daphnia magna, over an exposure period of 48 hours.

All validity criteria were met during this study.

Under the conditions of this Daphnia magna acute immobilisation study the observed endpoints for the effect of Reactive Red F08-0146 were the following:
Parameter Test item concentration [mg/L]
(48 hr) nominal measured
EC50 > 100 > 87.1
NOEC 100 87.1
LOEC > 100 > 87.1

Executive summary:

Acute toxicity of Reactive Red F08-0146 on Daphnia magna was assessed in an Acute Immobilisation Test, over an exposure period of 48 hours in a semi-static test system. The frequency of the water renewal periods were 24 hours.

Toxic effects were not observed during the preliminary concentration range-finding test therefore a limit test was performed in the main experiment using only one test concentration (100 mg/L nominal) and one control group.

The test design included four replicates at each test group (test item and control). Each group comprised twenty Daphnia, five in each of the four replicate vessels, each containing approximately 40 mL test dilution.

There was no immobilisation in 20 daphnids exposed to each group. In addition, no abnormal behaviour or appearance of test animals was detected.

The test item concentration was analytically determined at the start and at the end of each renewal period. Measured concentrations at the end of the renewal periods deviated more 20 per cent from the initial values, however geometric mean value of measured concentrations was within ±20 per cent of the nominal concentration, therefore biological results are reported separately based on nominal and measured value.

All validity criteria were met during this study.

Under the conditions of this Daphnia magna acute immobilisation study the observed endpoints for the effect of Reactive Red F08-0146 were the following:
