Skin irritation

A skin irritation GLP study according to OECD guideline 404 was conducted in 3 male New-Zealand White rabbits in order to evaluate the irritation and corrosion effects of dioctadecyl ether on the skin (EVIC-CEBA, 1997). In this study 0.5 g of dioctadecyl ether moistened with distilled water was applied to the clipped dorsal skin of the rabbits for 4 hours (residual test substance was removed at the end of the exposure period). Animals were observed periodically for dermal reactions including the corrosion reactions at 1, 24, 48 and 72 hours after patch removal. Dermal reactions were not observed at 1, 24, 48 and 72 hours after removal of the test substance, i.e., the scores for erythema and edema were 0 at all observation time points.

 

Eye irritation

An eye irritation GLP study according to OECD guideline 405 was conducted in 3 female New-Zealand White rabbits in order to evaluate the irritation and corrosion effects of dioctadecyl ether on the eyes (EVIC-CEBA, 1997). In this study 0.1 g of dioctadecyl ether was applied to the right eye of the rabbits. One hour after instillation of dioctadecyl ether, diffuse purple enanthemae with lacrimations (conjunctivae score: 2) was observed in all animals and slight chemosis (score 1) was observed in one animal. Iris congestion (score 1) was noticed in all animals. 24 hours after instillation, slight redness (score 1) remains visible in the three animals with the persistence of the slight chemosis (score 1) in one animal. The reversibility was complete within 48 hours.

 

Justification for classification or non-classification

Based on the results of the skin and eye irritation studies, dioctadecyl ether does not need to be classified according to Directive 67/548/EEC and EU Classification, Labelling and Packaging of Substances and Mixtures (CLP) Regulation (EC) No. 1272/2008.
