Sampling and analysis
- Analytical monitoring:
- no
Test solutions
- Vehicle:
- yes
- Details on test solutions:
- Quantities of stock solutions of the test substance in Analar acetone were added to triplicate sets of 140 ml glass flasks so that, when made up to 140 ml with a reconstituted fresh water, an approximately logarithmic series of concentrations ranging from 2—200 mg/L was produced. The final nominal concentrations were: 2, 4.3, 9.3, 20, 43, 93 and 200 mg/L.
Test organisms
- Test organisms (species):
- Daphnia magna
- Details on test organisms:
- Daphnia magna, less than 24h old, were taken from a culture in STL derived from a strain obtained (via ICI Brixham Laboratory) from I.R.Ch.A., France The D. magna were cultured in glass vessels (0.5 to 4 litre) containing reconstituted fresh water. The cultures were held in a temperature controlled room, 20 ± 2°C, under a 16 h light 8 h dark cycle. The cultures were fed concentrated Chiorella vulgaris 0.5 mg/L of active dried yeast daily.
Study design
- Test type:
- static
- Water media type:
- freshwater
- Limit test:
- no
- Total exposure duration:
- 48 h
- Post exposure observation period:
- None.
Test conditions
- Hardness:
- The total hardness of the water was 190 mg/L as CaCO3
- Test temperature:
- The temperature of the test solutions was 20 ± 2°C.
- pH:
- pH was 8.1—8.3
- Dissolved oxygen:
- Concentration of dissolved oxygen was 8.8—9.2 mg/L
- Salinity:
- No data
- Nominal and measured concentrations:
- 2, 4.3, 9.3, 20, 43, 93 and 200 mg/L nominal.
- Details on test conditions:
- A 48h static toxicity test was carried out without renewal of the test solutions. Quantities of stock solutions of 2,3-epoxypropyl neodecanoate in Analar acetone were added to triplicate sets of 140 ml glass flasks. Three flasks served as controls and received no test substance. The concentration of acetone in all test dishes, including the controls, was adjusted to 0.1 mL/L. Ten D. magna, less than 24h old, were allocated to each flask. In order to minimise the risk of D. magna becoming trapped at the surface, where at all concentrations tested, globules of test substance were visible, loosely fitting black paper caps were placed over the flasks to create a darkened zone which the D. inagna would avoid. After 24h and 48h the numbers of imiuobilised D. magna were counted and recorded. D. magna were considered to be immobile if, when the contents of the flask were briefly stirred they did not swim during a 10 second period of observation.

- Basis for effect:
- mobility
- Details on results:
- At 4.3 mg.L 30% of the Daphnia were immobilized by 48 hr of treatment. At test substance concentrations of 9.3 mg/L and greater immobilization was generally 100% after 48 hr of exposure.
- Reported statistics and error estimates:
- The the 48 hr EC50 was 4.8 mg/L with 95% confidence limits 4.0—5.7 mg/L.
Any other information on results incl. tables
Globules of undissolved test substance were present on the surface of the water at all test concentrations. The test substance was therefore not wholly soluble at any of the concentrations tested.
Applicant's summary and conclusion
- Validity criteria fulfilled:
- yes
- Conclusions:
- The 48 hr EC50 value for the test substance to Daphnia magna was estimated to be 4.8 mg/L.
- Executive summary:
The 48 hr EC50 of the test substance, 2,3-epoxypropyl neodecanoate to Daphnia magna was estimated by an O.E.C.D. test guideline 202 study. The 48 hr EC50 value for the test substance to Daphnia magna was estimated to be 4.8 mg/L. Therefore, it is anticipated that Neononanoic acid glycidyl ester will have a similar EC50 value under the conditions of the study.
