Description of key information

One eye and one skin irritation study were conducted with Sodium Ascorbyl Phosphate. In both studies only slight irritation scores were observed, which were reversible within the observation period of 72 hours.

Skin irritation:

In a primary dermal irritation study (BASF AG, 1997), six young adult New Zealand White Rabbits (3/sex) were dermally exposed to 0.5 g of moistened Sodium Ascorbyl Phosphate (85.4 %) for 4 hours. Animals then were observed for 3 days. Irritation was scored according to OECD 404, EU method B.4 and OPPTS 81-5.

Slight erythema were observed which were reversible within 72 h (24/48/72 h mean scores: 0.6 for erythema, 0.0 for edema).

Based on this study Sodium ascorbyl phosphate is evaluated as not skin irritating.

Eye irritation:

In a primary eye irritation study (BASF AG, 1997) 0.1 mL bulk volume (58 mg) of Sodium Ascorbyl Phosphate (85.4 %) was instilled into the conjunctival sac of the right eye of young adult new Zealand White rabbits (2 females, 4 males) for 24 hours.  The eye was washed with water after 24 hours. Animals then were observed for 3 days.  Irritation was scored according to OECD 404, EU method B.5 and OPPTS 81-4.

Slight conjunctivae redness and chemosis was observed which were reversible within 72 hours (24/48/72 h mean scores: 0.0 for cornea and for iris, 0.8 for conjunctival redness and 0.1 for chemosis).

In this study, Sodium Ascorbyl Phosphate is not an eye irritant.

Justification for classification or non-classification

Skin irritation:

Based on the results (24/48/72 h mean scores: 0.6 for erythema, 0.0 for edema) Sodium Ascorbyl Phosphate is classified as not irritating according to EU Directive 67/548/EC amended and repealed by EU Regulation No.1272/2008 and CLP.

Eye irritation:

Based on the results (24/48/72 h mean scores: 0.0 for cornea and for iris, 0.8 for conjunctival redness and 0.1 for chemosis) Sodium Ascorbyl Phosphate is classified as not irritating according to EU Directive 67/548/EC amended and repealed by EU Regulation No.1272/2008 and CLP.
