Analytical monitoring:
Details on test solutions:
To accelerate the solution procedure a five fold amount (30 mg/1) of the maximum water solubility of the test substance in the
preliminary test (6 mg/1) was weighed into water, treated for 60 seconds at 8000 rpm.with an ultra-turrax and afterwards stirred
an a magnetic stirrer for 24 hours. To remove undissolved particles, the resulting emulsion was filtered using a folded filter of the
pore size 7-12 gm.
Test organisms (species):
Danio rerio (previous name: Brachydanio rerio)
Details on test organisms:
- supplier: Bio International B.V. (Netherlands)
- body length: 2.5 - 3.5 cm
- medication: none
- mortalities during acclimatization: < 2 %
- keeping water: synthetic fresh water in accordance with ISO
Test type:
Water media type:
Limit test:
Total exposure duration:
96 h
total hardness 14.1 °dH (251.5 mg/l CaCO3)

Test temperature:
The temperature varied between 21.3 – 21.8 °C (daily monitoring)

pH-values varied betweeen 7.8 – 8.0 (daily monitoring)

Dissolved oxygen:
Oxygen concentration varied between 8.1 – 8.6 mg/l (daily monitoring)

- ventilated
- no feeding during test
- test vessel: glass aquarium, 5 l test media
- number of fish per test vessel: 10

Conc. based on:
other: determined as TOC and back-calculated to the parent compound
Basis for effect:
mortality (fish)
Remarks on result:
other: Arithmetic mean of analytical values
Details on results:
The above test result refers to a concentration of the test substance which was calculated directly from analytically determined TOC values.
Executive summary:

For fish (Danio rerio) a LC0 of >= 8.1 mg/l after 96 h was obtained for 4,4-methylenedicyclohexyl diisocyanate. The study was conducted according to Directive 92/69/EEC, C1 (Bayer AG, 2000). The result relates to the arithmetic mean of analytical values.

Description of key information

Acute toxicity to fish (Danio rerio) has been investigated in a limit test under static conditions according to Directive 92/69/EEC, C.1. A concentration of 30 mg/l 4,4’-methylenedicyclohexyl diisocyanate was pretreated before start as described above. A 96 h-LC50 of > 8.1 mg/l was observed (Bayer AG, 2000b). At this concentration level 10 % mortality was detected, however, in a range finding test no mortality occurred at the highest concentration tested of 6 mg/l (nominal).
