Test organisms

Test organisms (species):
Daphnia magna
Details on test organisms:
Daphnia magna Straus were obtained by aperiodic parthenogenesis from own laboratory breeding. Daphnia were fed by a mxture of algae. The sensitivity of test species is periodically monitored by testing with the reference substance, potassium dichromate. The grow-up gravid females are separeted into dilution water. Young daphnia born in 24 hours are segregated to the handling container from which they entered the experiment. At handling a care should be taken of not to penetrate the air under animals shell.

Results with reference substance (positive control):
verification test was carried out in period from 11.12.2007 to 13.12.2007
results:
24h - EC50=1.15 mg/L (95% CL)
48h - EC50= 0.80 mg/L (95% CL)
The result of last interlaboratory test arrangment by Centre for Assessment of Laboratories of T.G.Masaryk Water Research Institute Prague in 2006 for potassium dichromate was:
48-hour EC50= 0.61 - 1.11 mg/L
The immobilisation of test species in reference substance solution expressed as EC50 fits into the accepted range in the interlaboratory tests.
