Treatment of 3 male Sprague-Dawley rat with isophorone (150 mg/kg) by  gavage for 14 consecutive days resulted in a significant 

intensification  of a protein band corresponding to kidney-type-alpha2µ-globulin with a molecular mass calculated to be ~ 16kDa.

However, intraperitoneal treatment with leupeptin and E-64, well known cystein protease inhibitors, only slighthly increasing this kidney-type-alpha2µ-globulin band.

Isophorone form protein complexes with alpha2µ-globulin in vivo (SDS-page and immunoblotting after 14 days isophorone administration).

Effects of alpha2µ-globulin accumulating agents on alpha2µ-globulins in rat kidneys were examined by sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and immunoblotting analysis. The treatment of male animals with isophorone (150 mg/kg) by gavage for 14 consecutive daysresulted in a marked intensification of a protein band corresponding to kidney-type-alpha2µ-globulin, with a molecular mass calculated to be ~ 16 kDa.
