Description of key information

The long-term toxicity to aquatic invertebrates was determined by performing a semi static daphnia reproduction study in accordance with guidance OECD 211 and GLP. The daphnia were exposed to a concentration range of 0.6, 1.1, 1.9, 3.5, and 6.3 µg/L. Samples taken from stock solutions in DMSO used in the preparation of test solutions were analysed. The analysed concentrations ranged was 80 - 99% relative to the target concentrations, indicating proper preparation of the stocks. Based on these results, the effect parameters were based on analytically confirmed nominal concentrations. 


No mortality of the parental daphnids was observed during the test period in the blank control, the solvent control and all the test concentrations. On average, 174 and 156 offspring were produced per surviving daphnid in the blank and solvent control treatment, respectively. The number off offspring in all treated groups was similar to the solvent control treatment. Statistical analysis showed no statistically differences in the reproduction of the daphnids in all test concentrations.


No dose-related and statistically significant reduction of parental body length was found at the range of concentrations tested.



In conclusion, under the conditions of the present study Pigment Orange 5 did not affect reproduction, growth and survival of Daphnia magna at the highest test concentration of 6.3 μg/L after 21 days of exposure(NOEC).



 


All the acceptability criteria were met and therefore the study was considered to be valid.
