Materials and methods

Test guideline
Qualifier:
no guideline followed
Principles of method if other than guideline:
13 week study to evaluate the cumulative toxic effects of repeated administration of furfural and to determine the doses to be used in 2 year study. The publication also includes 16 day and 2 year studies. Only the 13 week study is reported in this summary and only the results relating to the reproductive organs are discussed. The data from the two year assay are not considered relevant for potential reproductive effects given the age of the animals at the time of evaluation.

Effect levels

Dose descriptor:
NOAEL
Effect level:
300 mg/kg bw/day (nominal)
Based on:
test mat.
Sex:
male/female
Basis for effect level:
other: no reported effects on the weight or histology of the reproductive organs at this lethal dose

Observed effects

The dose levels of 600 and 1200 mg/kg/day were lethal for the mouse. No effects on the weights or histology of the reproductive organs were reported at 300 mg/kg/day, a dose level that produced lower body weights and increased liver weight and liver histopathology.

Applicant's summary and conclusion

Conclusions:
13 weeks of oral administration of furfural at 300 mg/kg/day caused reduced body weight and increased liver weight in mice but no effects on the weights or histology of any of the reproductive organs were reported.
Executive summary:

The purpose of this 13 week study was to evaluate the cumulative toxic effects of repeated administration of furfural and to determine the doses to be used in a 2 year mouse study. The data from the two year assay are not considered relevant for potential reproductive effects given the age of the animals at the time of evaluation.

In the 13 week study, the dose levels of 600 and 1200 mg/kg/day were lethal to the mice. 300 mg/kg/day caused reduced body weight and increased liver weight but no effects on the weights or histology of any of the reproductive organs were reported.
