Reference
Endpoint:
short-term toxicity to fish
Type of information:
experimental study
Adequacy of study:
key study
Study period:
From 24 OKT 1980 to 31 OKT 1980
Reliability:
2 (reliable with restrictions)
Rationale for reliability incl. deficiencies:
guideline study without detailed documentation
Qualifier:
equivalent or similar to guideline
Guideline:
OECD Guideline 203 (Fish, Acute Toxicity Test)
GLP compliance:
no
Analytical monitoring:
no
Vehicle:
no
Details on test solutions:
The test material was solubilized in aliquots of test water and applied to the test basins. Even distribution was accomplished by stirring with a glass rod.
Test organisms (species):
Leuciscus idus melanotus
Details on test organisms:
- Origin of test organisms: Paul Eggers, 2345 Hohenweststedt
- Acclimatization: The fish were kept at minimum for 14 days before start of the test at 20 deg C in dechlorinated tap water.
- Body weight: 1.7 - 2.7 g (arithmetic mean = 2.1 g; SD = 0.28 g; n=10)
- Corpulence factor: 1.03 - 1.6 (arithmetic mean = 1.18; SD = 0.185; n=10)
- Feeding: Tetra Min (Tetra-Werke, Melle, Germany)
Test type:
static
Water media type:
freshwater
Limit test:
no
Total exposure duration:
96 h
Hardness:
Total hardness: 1,7 mM (Mg++ and Ca++);
Test temperature:
20 +/- 1 deg C
pH:
7.8 - 8.3
Dissolved oxygen:
> 8 mg/L
Nominal and measured concentrations:
Nominal test material concentrations:
0, 1, 10, 100 and 500 mg/L, corresponding to
0, 0.45, 4.5, 45 and 225 mg/L submission substance
Details on test conditions:
Test water:
De-ionized water, supplemented with the following salts:
- 192 mg/L NaHCO3
- 120mg/L CaSO4*2H2O
- 120 mg/L MgSO4
- 8 mg/L KCl

Test basins: Made from glass (40x25x30 cm) filled with 20 L test water. Test temperature was kept constant by means of a water bath.
Aeration: By glass capillary, 100 mL air / min;
Illumination: day-night regime 12 h day, 12 h night with approximately 700 Lux.

Directly before and directly after addition of test substance as well as after 2, 24, 72 and 96 hours pH, O2 and temperature was determined in each test basin. During tho whole experiment the fish were observed reularily and behaviour was recorded. At the end of the test fish were killed in 0.05 % Tricain (CAS 886-86-2), dissected and macroscopically examined.
Reference substance (positive control):
no
Duration:
96 h
Dose descriptor:
LC50
Effect conc.:
> 500 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Remarks:
500 mg/L test material corresponding to 225 mg/L submission substance.
Basis for effect:
mortality (fish)
Duration:
48 h
Dose descriptor:
LC50
Effect conc.:
> 500 mg/L
Nominal / measured:
nominal
Conc. based on:
test mat.
Remarks:
500 mg/L test material corresponding to 225 mg/L submission substance
Basis for effect:
mortality (fish)
Details on results:
During the test period of 96 hours in none fo the tested concentration groups any fish died. The LC50 for 48 and 96 hours is therefore above 500 mg/L test material concentration.
Behaviour of fish was normal for all test groups and no deviations from the control group could be observed.
Dissection of fish after termination of experiment resulted in no macroscopically determinable alterations.
pH (8.1 - 8.3) and O2-concentrations (8.5 - 8.8 mg/L) were in all test groups in the range of control values.
Validity criteria fulfilled:
yes
Remarks:
However no analytical monitoring. The submission substance is expected to be stable in water over the time period of the experiment.
Conclusions:
In an acute toxicity test on Leuciscus idus performed equivalent to OECD 203 (reliability category 2) the LC50 (48 and 96h) was determined as > 500 mg/L test material corresponding to 225 mg/L submission substance.
Executive summary:

In an acute toxicity test on Leuciscus idus performed equivalent to OECD 203 (reliability category 2) the LC50 (48 and 96h) was determined as > 500 mg/L test material corresponding to 225 mg/L submission substance.

During the test period of 96 hours in none of the tested concentration groups any fish died.

Description of key information

One acute toxicity studies of reliability 2 for Pigment Orange 13 on Golden orfe is available.
Under the conditions of this test the LC50 of the submission substance after 96 hours was > 500 mg/L (based an nominal concentration) and is above the solubility limit of the test substance in the test water (4.5 μg/L).

For all three pigments of the Diazopyrazolone group consistently no aquatic toxicity has been found up to nominal concentrations well above the solubility limit. Therefore it is justified to use the data for all members of the group.
