The potential comedogenic effect of 3,5,5-trimethylhexyl 3,5,5-trimethylhexanoate (CAS 59219-71-5) was evaluated in rabbits in a study performed using a test protocol based on Kligman, A.M. and Kwong T. (Brit J Dermatology 1979) and under GLP conditions (Bouffechoux, 1998). 0.1 mL of the undiluted test substance was applied to the internal base of one ear of 6 male rabbits for 5 days/week during 2 consecutive weeks. The other ear of each rabbit remained untreated and served as control. After exposure ended, the epidermis of the treated and control skin areas was separated from underlying tissue and stained with Sudan Red III. The tissue was mounted on slides and examined microscopically; whereby the skin integrity, number of follicles and number of comedones was assessed. The comedogenicity scores for the individual animals' treated ear were 1.24, 2.63, 4.40, 2.13, 4.95 and 1.07, respectively. The mean score was 2.72 (on a scale from 0 - > 7), indicating a slight comedogenic potential. For the control ears, all the scores were 0. No skin irritation was noted at the test sites at the end of the study period.
