Additional information

The acute toxicity of the test material to freshwater fish Oncorhynchus mykiss (rainbow trout), was determined in the key study by Mahabir and Glover (2002). The study was conducted in accordance with the standardised guideline OECD 203 under GLP conditions. The study was reported and performed to a high standard; however, there were minor deviations from the guideline which were not thought to affect the outcome of the results. As a result, the study was assigned a reliability score of 2 in accordance with the principles for assessing data quality as defined in Klimisch et al. (1997).

Fish were exposed to the test material for 96 hours under static conditions at nominal concentrations of 2.6, 6.4, 16, 40 and 100 mg/L, along with the control and solvent (DMF) control. The DMF was used as a solubilising agent due to the poor solubility of the test material.

There was 100 and 15 % mortality in the 100 and 40 mg/L dose groups, respectively. All control animals survived until study termination as did all animals in the 2.6, 6.4 and 16 mg/L dose groups.

After 24 hours, one fish in the 16 mg/L group was observed to have a complete loss of equilibrium. After 48 hours, some fish in the 40 mg/L dose group were observed to be lying on one side, respiring heavily. After 72 hours, some fish in the 16 and 40 mg/L dose groups were observed to be lethargic.

All fish in concentrations 6.4, 16 and 40 mg/L were dark in colour by day 1. All test animals in the 16 and 40 mg/L test chambers were also observed to have curved spines on day one, while some exposed to the 6.4 mg/L also had curved spines. By day 3, fish in the 6.4 mg/L test chambers had returned to normal colouration and curved spines could no longer be detected.

Under the conditions of the test, the 96 hour LC50 in Oncorhynchus mykiss (rainbow trout) was determined to be 55 mg/L (95 % CL 48 to 64 mg/L) with a NOEC of 6.4 mg/L.
