Sampling and analysis

Analytical monitoring:
yes
Details on sampling:
At the start and end of the test, the pH values, dissolved oxygen concentrations and water temperature were determined in each treatment. In addition, the appearance of the test media was visually controlled and documented at the start of the test, after 24 hours and at the end of the test.

Test solutions

Vehicle:
no
Details on test solutions:
The following test concentrations of Methyl Octine Carbonate were tested: The dilutions 1:220, 1:100, 1:46, 1:22 and 1:10 of a stock solution with a loading rate of 100 mg/L. Additionally, a control (test water without test item) was tested in parallel.

Test organisms

Test organisms (species):
Daphnia magna
Details on test organisms:
The study was performed with young daphnids of the species Daphnia magna Straus. A clone of this species (originally from the Daphnia Collection of the University of Basel/Switzerland in 2015) is successfully bred in IES Ltd Laboratories under temperature and light conditions identical to those of the tests.

Study design

Test type:
static
Water media type:
freshwater
Limit test:
yes
Total exposure duration:
48 h

Test conditions

Test temperature:
The water temperature during the test was maintained at 21 °C.
pH:
The pH value of the test media was in the range of 7.6 to 7.9
Dissolved oxygen:
At the beginning and end of the test period, the dissolved oxygen concentrations in the test media and control were at least 7.9 mg/L.
Nominal and measured concentrations:
Nominal concentrations: 1:220, 1:100, 1:46, 1:22 and 1:10 dilutions of an equilibrated test item solution with a loading rate of 100 mg/L
Mean measured concentrations: corresponding to the above dilutions were respectively, n.a., 0.38, 0.8, 1.8 and 4.1 mg/L
n.a. = not analyzed since below the NOEC determined in this test
Details on test conditions:
The test was performed in a temperature-controlled room with continuous monitoring of the room temperature. The test water temperature was maintained at 21 °C.
A 16-hour light to 8-hour dark cycle with a 30-minute transition period was used. Light intensity during the light period was approximately between 15 and 18 µmol m-2 s-1.
The daphnids were not fed during the test.
Reference substance (positive control):
not required

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Executive summary:

The test item Methyl Octine Carbonate had acute toxic effects on Daphnia magna in a static 48-hour test. The 48-hour EC50was calculated to be 1.1 mg/L with 95%-confidence limits of 0.88 and 1.5 mg/L (based on mean measured concentrations). The 48-hour NOEC of Methyl Octine Carbonate was 0.38 mg/L, since no toxic effect was observed up to and including this test concentration.
