In different studies, Butane-1, 2, 3, 4-tetracarboxylic acid (1703-58-8) has been investigated for potential of skin sensitization to a greater or lesser extent. The studies are based on in vivo experiments in human for target chemical Butane-1, 2, 3, 4-tetracarboxylic acid (1703-58-8) and structural read across. The read across substances share high similarity in structure and log kow .Therefore, it is acceptable to derive information on skin sensitization from the analogue substance.
The skin sensitization potential of Butane-1, 2, 3, 4-tetracarboxylic acid (1703-58-8) was estimated by SSS (2017) using OECD QSAR toolbox v 3.3 with log kow as the primary descriptor and considering the six closest read across substances. Butane-1, 2, 3, 4-tetracarboxylic acid (1703-58-8) was predicted to be non-sensitizing to the skin of female CBA mouse.
Supported to the above weight of evidence a study on read across chemical Citric acid from Cosmetic Ingredient Review, 2012, skin sensitization test was performed in 56 patients by applying 4% in a cuticle cream by HRIPT methods. Semi occlusive patches were applied 3x/wk for 3 wks in induction exposure and continue after 2 week in challenge exposure. After the experiment, no reaction was obtain hence, the citric acid was considered to non-toxic to human patient and based on that it can be considered to be non-sensitizer.
Another read across Dodecanedioic acid (693-23-2) from IUCLID dataset, 2000, skin sensitization test was performed using Guinea pig maximization test in 20 animals for 24 or 48 hrs according to OECD guideline 406. Patch test was performed and after 24 and 48 hrs the Dodecanedioic acid (693-23-2) was considered to be non-sensitizing after exposure to Guinea pig.
Thus based on the above weight of evidences, it can be considered that Butane-1, 2, 3, 4-tetracarboxylic acid (1703-58-8) was not a skin sensitizer supported by the majority of the above studies. Thus comparing the above annotations with the criteria of CLP regulation, Butane-1, 2, 3, 4-tetracarboxylic acid (1703-58-8) can be considered as not classified for skin sensitization.
