One key study along with three supporting studies were identified for ethylene glycol hexyl ether (EGHE) acute toxicity to fish. In a 96 -hour Static Acute Fish test (identified as the key study) with fathead minnows (Pimephales promelas), conducted in accordance with OECD TG 203, EPA OTS 797.1400 and ASTM guidelines, the 96 hour LC50 value of EGHE was found to be 140 mg/L, with 95% confidence limits that ranged from 130 -140 mg/L and an NOEC value of 40 mg/L. In another 96 -hour Static Acute Fish test on fathead minnows (Pimephales promelas), conducted in accordance with the EPA and ASTM guidelines, the 96 hour LC50 value of EGHE was also reported to be 140 mg/L. Similarly, in an additional 96 -hour Static Acute Fish test on fathead minnows (Pimephales promelas), conducted in accordance with the EPA and ASTM guidelines, the 96 hour LC50 value of EGHE was found to be 139.5 mg/L. A fourth supporting study, following OECD guidelines (OECD 203) and GLPs, reported a 96 hour LC50 value for zebrafish as >100 - 220 mg/L.

One key study and three supporting studies were identified for the acute toxicity of ethylene glycol hexyl ether (EGHE) to aquatic invertebrates. A key study that evalauted the acute toxicity of EGHE to the cladoceran Daphnia magna (Klimisch score of 2) reported a 48 hour EC50 of 145 mg/L, which was also reported by one of the supporting studies. Two additional studies (Klimisch scores of 2) conducted in accordance with EPA/ASTM guidelined with Daphnia magna reported 48 hour LC50 values of 305 mg/L.

The toxicity of ethylene glycol hexyl ether (EGHE) was evaluated in an algal study with Desmodesmus subspicatus (formerly Scenedesmus subspicatus) following DIN 38412, part L9 guidelines and given a Klimisch rating of 2. The study reported a 72-hr EC50 value of 198. mg/L (growth rate) and a 72-hr EC10 of 23.4 mg/L (growth rate).
