According to the OECD guideline 471 a biological relevant mutagenic potential of the test item was determined under the conditions of the test without and with metabolic activation.
Executive summary:

The test item Basic Blue 99 was assessed for the potential to induce gene mutations in the Salmonella typhimurium reverse mutation test. The study was performed according to OECD guideline 471. Bacterial strains Salmonella typhimurium TA98, TA100, TA102, TA1535, TA1537 were used in the study. Exposure of the test system has been performed using the plate incorporation method. The test item was applied at concentrations of 1, 0.316, 0.1, 0.0316, 0.01 and 0.00316 mg/plate (main test) with and without metabolic activation (S9 rat liver (2mg/mL)). Depending on the bacterial strain, precipitates and/or cytotoxicity was observed in higher concentrations. According to the OECD guideline 471 the tested concentration range was justified. The respective solvent control produced a low number of revertants. The strain specific positive controls for TA98, TA100, TA102 and TA1535 were shown to produce the expected increase in revertant colonies, with and without metabolic activation. For the strains TA98, TA100 and TA1537 a mutagenic potential of the test item (with and without metabolic activation) was determined. According to the OECD guideline 471 a biological relevant mutagenic potential of the test item was determined under the conditions of the test without and with metabolic activation.
