- Details on test solutions:
- A stock solution was prepared by weighing out 0.5133 g test substance, then made it up to 100 mL with TweenⓇ 80 (0.5 g) to give 5000 mg/L.
0.100, 0.250, 0.626, 1.562, 3.906 and 9.766 mL of stock solution were transferred into 10 mL of volumetric flasks and filled up to the 10 mL marker with TweenⓇ 80 : ground water (0.5:99.5, w/v) in test water to prepare working solutions.
5 mL of each working solution was added test water and filled up to the 5 L marker to prepare test solutions.
Solvent concentration (Tween(R) 80) of each test solution was 50 mg/L.
The range-finding test was performed in 0.1, 1.0, 10.0 and 100.0 mg/L. The inhibition rate for the average specific growth rate was –2.1, -8.8, 64.2 and 97.0 %, respectively.
The inhibition rate for the yield was 13.4, 18.0, 90.8 and 99.6 %, respectively.
The definitive test was performed in 0.50, 1.25, 3.13, 7.81, 19.53 and 48.83 mg/L (nominal concentration, geometric series 2.5).
As analysis result of test substance in test solution, the measured mean concentrations were 0.42, 1.02, 2.38, 6.09, 14.48 and 41.00 mg/L, respectively.
Negative control : OECD medium was used for negative control.
Solvent control : 50 mg/L of TweenⓇ80 (in OCED medium) was used for solvent control. - Test organisms (species):
- Raphidocelis subcapitata (previous names: Pseudokirchneriella subcapitata, Selenastrum capricornutum)
- Details on test organisms:
- Freshwater alga (Pseudokirchneriella subcapitata , ATCC22662).
The culturing vessels were shaken and placed in the culturing apparatus (J-SCI, Jisico Co., Korea). The cultures were maintained at a temperature in the range of 21oC - 24oC. The surface of cultures were received continuous, uniform fluorescent illumination in the range of 4440 Lux - 8880 Lux. The culturing vessels were 250 mL of glass flasks.
The temperature in the incubator was recorded automatically every 1 h using TR-71U thermo-recorder (TND Co., Japan). The light intensities were measured in the incubator using ANA-11 digital lux meter (Tokyo Koden Co., Japan) twice a week. As the results of the monitoring, there was not observed an alteration effecting the test.
The growth medium was prepared by OECD medium (OECD TG 201, Annex 3 Growth media, 2006-03-23). The pH values of OECD medium was measured using PP-15 pH meter (Sartorius AG, Germany). - Test type:
- static
- Water media type:
- freshwater
- Limit test:
- no
- Total exposure duration:
- 96 h
- Test temperature:
- The mean temperature in the incubator was 22.7oC (min. 22.1oC - max. 23.6oC)
- pH:
- mean pH was 8.22 ± 0.30 (min. 7.84 - max. 8.60)
- Nominal and measured concentrations:
- The definitive test was performed with 0.50, 1.25, 3.13, 7.81, 19.53 and 48.83 mg/L (nominal concentration, geometric series 2.5). The analysis of test concentrations were performed at the start of the test and the end of the test.
The analysis results of test substance in the test solution was 0.42, 1.02, 2.38, 6.09, 14.48 and 41.00 mg/L. - Details on test conditions:
- The pre-culture was incubated under the test condition and used when exponentially growing, after incubation period of 4 days. The pre-cultured alga was inoculated (1 ×
10^4) cells/mL in each vessel. And each test concentration has 3 replicates. The test vessels were placed in the shaking incubator (J-SCI, Jisico Co., Korea) maintained at the temperature of 21oC - 24oC, the light intensity of 4440 Lux - 8880 Lux, always light and 100 rpm. The test vessels were 250 mL of glass flasks. - Reference substance (positive control):
