Administration / exposure

Details on inhalation exposure:

Details on study design:
Exposure type: Saturated atmosphere of chemical vapor:

Substantially saturated vapor was prepared by spreading 50-100 grams of 1-naphthol over a 200 cm2 area on a shallow tray placed near the top of a 120 liter plexiglas chamber which is then sealed for at least 16 hours while an intermittently operated fan agitates the internal chamber atmosphere. Rats were then introduced into a gasketed drawer-type cage designed and operated to minimize vapor loss. Animals were exposed for six (6) hours and observed for mortality and toxicity for 14 days.

Exposure type: Saturated atmosphere of mist, vapor and decomposition products:

1-naphthol was heated to 179 degree C. Air was passed over the heated sample and mist, vapor, and any oxidation products generated were delivered to rats in a 9 liter glass exposure chamber. Animals were exposed for six (6) hours. Animals were observed for mortality and toxicity for 14 days.

Applicant's summary and conclusion

Interpretation of results:
study cannot be used for classification
Remarks:
STOT SE Cat 3 (Note, this is also a harmonized classification)
Conclusions:
LC50 not determinable as the concentrations were not provided. However, STOT SE Cat 3 was assigned per UN GHS due to the eye irritation and hypoactivity from exposure to heated saturated vapor. No mortality reported.
Executive summary:

Rats were exposed for 6 hours with saturated atmosphere of 1 -naphthol or a saturated atmosphere of mist, vapor, and decomposition products of heated (179 degrees C) 1 -naphthol. The LC50 was not reported and the exposure concentrations were not provided. No deaths occured. The results cannot be used for Acute Toxicity - inhalation classification. However, STOT SE Cat 3 was assigned per UN GHS due to the hypoactivity from exposure to heated saturated vapor of 1 -naphthol.
