All loading levels and the control were analytically verified via GC-MS after 0 (start of exposure), 24 and 72 hours (end of the
exposure). Separate replicates for the test item analysis after 0, 24 and 72 hours were prepared with algae at the start of the exposure and incubated under test conditions until analysis.

Five water accommodated fractions (WAF) were prepared with a
accommodated fraction nominal loading of the test item of 6.25 - 12.5 - 25.0 - 50.0 – 100 mg/L (spacing factor 2). A slow stirring procedure with a minimized headspace was applied. Gentle stirring (to avoid formation of an emulsion) was carried out with a magnetic stirrer at room temperature for 24 ± 1 hour. After a separation phase of four hours at room temperature the WAFs were taken from the bottom of the flask and were used for testing. Each WAF was checked via laser beam (Tyndall effect) for undissolved test item (formation of an emulsion). The resulting water accommodated fractions (WAF) were used in the test. Additionally, the loading level 3.125 mg/L was tested. Therefore, the WAF with a loading of 6.25 mg/L was diluted with dilution water. The loading levels are based on the results of the first test.

Per definition of the WAF, all terms related to concentration level were given as loading level because partly dissolved compounds and mixtures cannot be related to concentrations, therefore, all effect values given are based on the nominal test item loadings of CEDRENE WASHED.
Details on test conditions:
The study was conducted under static conditions with an initial cell density of 6839 cells/mL. Since the test item is an UVCB five water accommodated fractions (WAF) with the nominal test item loadings 3.13 - 6.25 - 12.5 - 25.0 - 50.0 - 100 mg/L (spacing factor of 2) were prepared with dilution water and slowly stirred for 24 hours (20 rpm, room temperature). Three replicates were tested for each test item loading and six replicates for the control. The environmental conditions were within the acceptable limits.

Details on results:
The NOEL and LOEL were determined by calculation of statistical significant differences of growth rates and yield. As a standard, One Way Analysis of Variance (ANOVA) and DUNNETT’s test were used for NOEL/LOEL calculations.
Results with reference substance (positive control):
The toxicity of potassium dichromate (Sigma, batch number MKBV0900V, purity 99.0 %, CAS RN 7778-50-9) to the unicellular freshwater green alga Pseudokirchneriella subcapitata was determined over a period of 72 hours from 2017-10-10 to 2017-10-13 (with headspace) and 2017- 10-09 to 2017-10-12 (without headspace), respectively. The reference item toxicity is in the valid range following test facility SOPs.
