Test material

Method

Target gene:
In the Salmonella typhimurium strains (TA 1535, TA 100, TA 1537, TA 98) the amino acid histidine locus is the target gene.
Species / strain
Species / strain / cell type:
S. typhimurium TA 1535, TA 1537, TA 98 and TA 100
Metabolic activation:
with and without
Metabolic activation system:
Rat liver S-9 mix
Test concentrations with justification for top dose:
10, 50, 100 and 500 µg/plate for TA 1535, TA 1537 and TA 98
10, 50, 10, 200 and 300 µg/plate for TA 100 because 500 µg/plate proved already to be toxic to this strain.

Results and discussion

Test resultsopen allclose all
Species / strain:
S. typhimurium TA 1535
Metabolic activation:
not specified
Genotoxicity:
negative
Species / strain:
S. typhimurium TA 1537
Metabolic activation:
not specified
Genotoxicity:
negative
Species / strain:
S. typhimurium TA 98
Metabolic activation:
not specified
Genotoxicity:
negative
Species / strain:
S. typhimurium TA 100
Metabolic activation:
with and without
Genotoxicity:
positive
Additional information on results:
TA 100 responded with a concentration-dependent increase in revertants with and without metabolic activation. With metabolic activation the effect on TA 100 was significantly enhanced.

Applicant's summary and conclusion

Conclusions:
It can be stated that 1,2-Bis(chloromethyl)benzene is mutagenic under the conditions of the applied bacterial test system.
