- Details on sampling:
- Range-finding test:
> concentrations: 123 mg Aldolase/l
> Frequency: at t=0 h and t=72 h
> Volume: 10 ml
> Storage: Not applicable, samples were analysed on the day of sampling
Final tests:
> concentrations: samples were taken from all concentrations and the blank-control

> Volume: 10 ml
> Storage: Not applicable, samples were analysed on the day of sampling - Vehicle:
- no
- Details on test solutions:
- Since the test concentrations should be based on protein concentrations, the test solutions were prepared using a stock solution of 1230 mg/l to obtain a concentration of 100 mg protein per liter. The stock solution was magnetically stirred for 15 minutes to accelerate the dissolving of the test substance in the test medium. The lower test concentrations were prepared by subsequent dilutions of the stock in test medium. The final test solutions were all clear and colourless.
- Test organisms (species):
- Raphidocelis subcapitata (previous names: Pseudokirchneriella subcapitata, Selenastrum capricornutum)
- Details on test organisms:
- TEST ORGANISM
- Common name: Selenastrum capricornutum
- Strain: NlVA CHL 1
- Source: In-house laboratory culture
- Age of inoculum (at test initiation): 3 or 4 days before the start of the test, cells from the algal stock culture were inoculated in culture medium at a cell density of 2.10E4 cells/ml. The pre-culture was maintained under the same conditions as used in the test.
- Method of cultivation: Algae stock cultures were started by inoculating growth medium with algal cells from a pure culture on agar. The suspensions were continuously aerated and exposed to light in a climate room at a temperature of 23 +/- 2°C.

- Test temperature:
- - first EC50 test: between 23.6 and 23.9°C
- final EC50 test: between 23.7 and 24.3°C - pH:
- - first EC50 test: The pH was generally within the limits prescribed by the protocol (6.0-9.0, preferably not varying by more than 1.5 unit). The pH of the blank-control and the lowest test concentration exceeded the upper limit at the end of the test due to a relatively high algal growth rate.
- final EC50 test: The pH of the blank control and all test concentrations exceeded the upper limit at the end of the test and some of the pH values also varied by more than 1.5 unit during the test period. This was due to a relatively high algal growth rate. - Dissolved oxygen:
