PREPARATION OF TEST SOLUTIONS
Range-finding test: A stock solution of 1000 mg/L was prepared by mixing 101.3 mg of the test substance in 100 mL of filtered nutrient solution. The solution was then placed on a magnetic stir plate and agitated for just over 4 hours.
Definitive test: A 10 mg/L stock solution was prepared, by mixing 10.1 mg of the test substance in 1000 mL filtered nutrient solution and stirring for 10 minutes.

Exposure concentrations one order of magnitude apart, were used to determine the range of concentrations required for definitive testing with S. capricornutum. the highest concentration tested was 1,000 mg/L, together with four lower concentrations (i.e. 100 mg/l, 10 mg/L, 1 mg/L, 0.1 mg/L) and a control (100% nutrient solution). S. capricornutum cells (1*10^4 cells/mL +- 1000 cells/mL) were added to each flask (1 replicate per concentration). As per test method, the exposure duration was 96 hours, with daily cell counts.

Definitive testing
A definitive multi-concentration EC50 test was conducted with nominal concentrations of 0.625, 1.25, 2.5, 5 and 10 mg/L plus controls. Each concentration was replicated 5 times. Observations of cell number and appearance were conducted, using a haemocytometer, on a daily basis. After 72 hours, the pH in one replicate sample was measured and the sample was sent for analysis, leaving four replicated remaining for the 96-hour counts. Changes in cell development or appearance, such as cell clumping, cell morphology, cell colour, cell shape, cell size etc. were reported.

The ecotoxicological evaluation of the effects of 4,4-dimethyl-oxazolidine (CAS 51200-87-4) indicated that the 72 and 96 -hour EC50s for cell number were 3.79 and 5.42 mg/L, respectively. The 72 and 96 -hour EC50s for area under the growth curve were 3.76 and 4.20 mg/L, respectively. The 0 to 72 and 0 to 96 -hour EC50s for growth rate were 6.57 and 7.14 mg/L, respectively. The NOEC and LOEC for 72 -hour cell number and area under the growth curve were 1.25 and 2.50 mg/L, respectively. For the other four endpoint parameters, the NOEC and LOEC were 2.50 and 5.00 mg/L, respectively.
Executive summary:

Ecotoxicological testing of Bioban CS-1135 (CAS 51200 -87 -4) was conducted to determine the growth inhibition of the freshwater green alga, Selenastrum capricornutum Printz, according to the OECD Method 201 (Alga, Growth Inhibition Test, 1984) and in general accordance with OPPTS Method 850.5400 (Algal Toxicity, Tiers I and II, 1996).

Results from the range-finding test indicated that the 50% inhibitory effect level and the no effect level for growth inhibition of Selenastrum capricornutum were between 1 and 10 mg/L for all endpoint parameters (72 and 96 -hour cell number, area under growth curve and growth rate).

The available analytical methods for the test item were not able to extract and quantify this compound. Calculations of all statistical endpoints for the definitive test were conducted using nominal concentrations. The 72 and 96 -hour EC50s for cell number were 3.79 and 5.42 mg/L, respectively. The 72 and 96 -hour EC50s for area under the growth curve were 3.76 and 4.20 mg/L, respectively. The 0 to 72 and 0 to 96 -hour EC50s for growth rate were 6.57 and 7.14 mg/L, respectively.
