Sampling and analysis
- Analytical monitoring:
- yes
- Details on sampling:
- Water samples were collected from freshly prepared batch (or pooled replicate samples as in
the Rainbow trout) of test solution for each treatment group and control at test initiation. All samples
were collected from mid-depth in the test solutions, placed in glass vials, and processed immediately
for analysis.
Test solutions
- Vehicle:
- yes
- Details on test solutions:
- A primary stock solution was prepared in well water at a nominal concentration of
5.0 mg/L. The stock was stirred with two top-down electric mixers for ~30 minutes to mix.
The primary stock solution appeared clear and colorless. Proportional aliquots of the primary
stock solution were diluted with U.V. sterilized well water to prepare the 0.063, 0.13, 0.25,
0.50, 1.0 and 2.0 mg/L test solutions. The test solutions appeared clear and colorless. All
organisms were transferred from old test solutions to freshly prepared test solutions at 48
hours.
For the rainbow trout study samples of freshwater were fortified at 0.600, 0.400 and 2.00
mg/L using a stock solution of Maropitant in methanol, and were analyzed concurrently with the test
samples. The measured concentrations for the matrix fortification samples ranged from 84.4 to 115%
of fortified concentrations.
Test organisms
- Test organisms (species):
- Oncorhynchus mykiss (previous name: Salmo gairdneri)
- Details on test organisms:
- Source: Thomas Fish Company, Anderson, California
Age at Test Initiation: Juveniles
Feeding: During holding, fed a commercially prepared diet supplied by Zeigler
Brothers, Inc., Gardners, Pennsylvania. No feeding during the test.

TEST MEDIUM / WATER PARAMETERS
- Source/preparation of dilution water: Water used for culturing and testing of the daphnids was obtained from a well located on the Wildlife International Ltd. site.
- Well water characteristics
(Mean of 4 weekly measurements taken prior to test initiation): Hardness
(135 mg/L as CaCO3,); Alkalinity (181 mg/L as CaCO3); Conductivity (301
µmhos/cm) and pH (8.2).
- Intervals of water quality measurement: DO at 0, 24, 48 (old and new solutions), 72 and 96 hours; Temperature and pH at test initiation, 48 hours (old and new solutions) and termination;
