In order to evaluate the aquatic toxicity of alkylation and transalkylation products of biphenyl with propene, short-term tests with fish (Bussard/ABC 1998a), daphnia (Bussard/ABC 1998b and Aniol/STZ 2011a) and algae (Bussard/ABC 1998 and Aniol/STZ 2011b) have been performed. In addition, long-term toxicity was determined in a 21 day daphnia immobilisation and reproduction test (Bussard/ABC 1998). All tests were conducted under GLP conditions according to OECD or EU test guidelines. Common problem was the low water solubility of the test substance.

In the tests of Bussard with fish and daphnia flow through systems were used. The daphnia test of Aniol and the Bussard algae test applied static conditions. In all tests of Bussard, nominal concentration was 12 µg/L, the water solubility of the substance determined in a preceding test. In the test of Aniol, a water accommodated fraction was used (nominal load 100 mg/L, no indication of undissolved material in the test solution). In the studies of Bussard, nominal concentrations in test solutions could not be confirmed to equal the nominal level. Measured concentrations were always lower. In the Aniol study, test concentration was below limit of quantification. In all tests, no adverse effects on test organisms were observed.

Differences in the toxicity values of Bussard do not demonstrate that any biological effects were noted. In all of the tests, no adverse effects were observed. The variable concentrations are related to the difficulty to force the test substance into solution. Even though in all flow-through tests the same diluter system was employed, the resulting measured concentrations were quite diverse. Toxicity thresholds could not be established. But the data derived from the tests indicate that there will be no aqueous toxicity in the range of the water solubility of the substance. This could be confirmed in acute screening tests with daphnia and alga (STZ 2011a,b) using Water-Accommodated Fractions (WAF) from loadings of 100 mg/L test substance without producing toxic effects.
