Propionyl chloride was tested in the Ames reverse mutation assay (GLP compliant study according to OECD guidelines 471 and 472) using Salmonella typhimurium strains TA98, TA100, TA1535 and TA1537 and Escherichia coli WP2 uvr A at 15.625 to 5000 µg/plate (standard plate test) and 15.625 to 250 µg/plate (preincubation test) with and without metabolic activation. Propionyl chloride was cytotoxic from 500 µg/plate onward in the standard plate test and at 125 µg - 250 μg/plate in the preincubation test. No precipitation of the test substance was found at any test concentration. Under the conditions tested, propionyl chloride was not mutagenic in any of the S. typhimurium strains and E. coli WP2 uvr A.

Propionyl chloride did not cause gene mutations in Salmonella typhimurium and Escherichia coli (Ames test).

Justification for classification or non-classification

Based on the results of the Ames test, propionyl chloride does not need to be classified according to Directive 67/548/EEC and according to the EU Classification, Labelling and Packaging of Substances and Mixtures (CLP) Regulation (EC) No. 1272/2008.
