Description of key information

In a GLP guideline study according to OECD 404 no skin irritating potential of 2-Hydroxy-4,6-diphenyl-1,3,5 -triazin was observed in New Zealand White rabbits. In a further GLP guideline study according to OECD 405 no eye irritating potential of 2-Hydroxy-4,6-diphenyl-1,3,5 -triazin was observed in New Zealand White rabbits.

Skin irritation:

A GLP guideline study (according to OECD 404, adopted 1981) is available, where 3 female New Zealand White rabbits were tested for the skin irritating potential of 2-Hydroxy-4,6-diphenyl-l,3,5 -triazin. The animals were treated for 4 h with 0.5 g of the moistened test substance on a gauze patch, which was loosely covered with aluminium foil. After 1, 24, 48, and 72 h after patch removal the skin of the animals was scored for erythema and edema.

No findings were observed at the side of the test patches in any animal. After 1 h an erythema score of 1 was observed in 2/3 animals. These findings were fully reversible within 24 h. No scores higher than 0 for erythema and edema were observed in any animal at the time points 24, 48, and 72 h. After 72 h the test was ended because no skin reactions have been observed. Thus, 2 -Hydroxy-4,6 -diphenyl-l, 3,5 -triazin is not irritating to the skin under the test conditions used.

Eye irritation:

A study to assess the eye irritating potential of 2-Hydroxy-4,6-diphenyl-1,3,5-triazin was conducted according to OECD TG 405 and GLP. About 55 mg of the test substance was instilled into the left eye of one male and two female New Zealand White rabbits. No rinsing of the eyes was performed and the scores were observed after the Draize scheme 1, 24, 48, and 72 h after beginning of the treatment. The untreated right eye served as a control. No effects in the untreated eyes of the rabbits were observed. No effects on cornea or iris were observed in the treated eyes of the three animals after 1, 24, 48, and 72 h. After 1 h redness (up to score 2) and chemosis (up to score 1) were observed. After 24 h again redness (up to score 1) and chemosis (up to score 1) were observed. The effects were fully reversible within 48 h after beginning of the treatment. Thus, 2 -Hydroxy-4,6 -diphenyl-1,3,5 -triazin is not irritating to the eye.

Respiratory irritation:

There exists no standard test method for the assessment of respiratory irritation. Thus, findings for skin/eye irritation and sensitisation are reviewed. There is no evidence that the test substance possesses a potential for respiratory irritation, because of the negative findings in skin/eye irritation and sensitisation studies that were performed.
