PREPARATION OF THE STOCK SOLUTION AND DILUTIONS
- For the preparation of the stock solution (12500 mg/l) the test substance was stirred for about 20 hours at about 22°C (Ikamag RCT basic, step 8) in demineralized water.
- The undissolved test substance was removed by centrifugation of the stock solution (20 min at 7000g
(HERAEUS Cryofuge 6-6, 6000 rpm)). The test was performed with the supernatant (eluate).

The following nominal concentrations were made by diluting the aqueous extract with demineralized water:

INOCULUM
- The test strain of Pseudomonas putida DSM 50026 used is obtained in regular intervais from DSM (German Collection of Microorganisms / Deutsche Sammlung von Mikroorganismen in Braunschweig) and is kept on agar slants for further cultivation in the Labor für Okotoxikologie of BASF AG Ludwigshafen.
- Pseudomonas putida is a gram-negative, aerobic, motile rod (length 2,0 to 4,0 pm , diameter 0,7 to 1,1 pm) with polar flagella. lt occurs ubiquitously in soil, surface water and waste-water. The optimal temperature for growth is among 25 and 30°C.
