The test was conducted according the the test guideline OECD Guideline 209 (Activated Sludge, Respiration Inhibition Test) (ISO 8192-1986). The activated sludge taken from a laboratory culture was fed with a synthetic sewage solution. The test item was exposed to the activated sludge for 3 hours.
First a range test with the test item concentrations of 100, 1000, 10000 mg/L was conducted. The range test resulted in a 8.3 % inhibition and a respiration rate of 17 mg/L*h at the concentration of 1000 mg/L. Therefore, the main test concentrations were set as follows: 1000, 1800, 3200, 5600, 10000 mg/L. The maximum test concentration was chosen based on the solubility of the test substance. Additionally, a control (water) was measured at the start and at the end of the exposure time and a reference substance (3,5-Dichlorophenol) with two different concentrations was also included. During the exposure time the oxygen concentration, growth inhibition, pH and temperature was measured.
The EC values were calculated with a graphic evaluation (Probit Analysis). The test resulted in an EC50 of 3534 mg/L and an EC10 of 894 mg/L for the test item Incozol 4.
