PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method:
For preparation of the 1 - 5.6 mg/L concentrations 0.02 mL Tween 80 were added to the test item and filled up with water. After sonification for 30 min and stirring for 5 min the solution was filled up to 200 mL end volume with water
.The 10 to 180 mg/L test concentrations were directly weighed in the test vessels and moistened with 0.095 mL Tween 80/L. Afterwards 200 mL water was added and the suspensions were homogenised by sonication and ultra-turrax.

At test item concentration above 10 mg/L the solutions were colored yellow. The measured water quality parameters (i.e. pH, oxygen concentration and temperature) as well as the analysed test item concentrations were within acceptable ranges.

Based on the nominal (and measured) concentrations of the test item, the 48-hour EC50 for Daphnia magna was 7.72 mg/L.
Executive summary:

In the acute immobilization test with Daphnia magna (STRAUS), the effects of the test item were determined at the test facility according to OECD 202 (1994).

The study was conducted under static conditions over a period of 48 hours with a test item solution with the nominal concentrations of 1, 1.8, 3.2, 5.6, 10, 18, 32, 56, 100 and 180 mg/L of the test item.

At test item concentration above 10 mg/L the solutions were colored yellow. Ten daphnids were exposed each concentration level and the control.

The concentrations of the test item were analytically verified via photometry in fresh media at the start (0 hours) and in old media at the end of the exposure (48 hours) in each concentration and in the control.

The measured concentrations of the test item were in the range of 89 to 111% of the nominal value.

The measured test item concentrations remained stable within ± 20% of the nominal limit concentration of the test item over the test period.

The validity criteria of the test guideline were fulfilled.
