A single exposure concentration was performed in the definitive study based on the Following :
Overview of Rational for Using a Single Exposure Concentration :
a) SCENTAURUS BERRY is a non-polar organic substance and is expected to behave and bioaccumulate in a linear fashion across a broad range of environmentally relevant exposure concentrations.
b) No concentration effects were expected because the test concentration (2 µg/L) was much lower than its water solubility (0.0567 mg/L), and, no acute toxic effect was observed in fish toxicity studies (96 hr LC50 >2 mg/L for Medaka).
c) Low BCF values were observed in the pre-study with common carp (< 100 L kg-1 wet wt), and, the BCF values observed at the two exposure levels in the pre-study were equivalent.
d) Reduction in the number of vertebrate animals employed in testing, conform with good animal welfare practice.

Details on results:
The concentration in test water at concentration level was close to the nominal concentration, and the variation of the test substance concentrations in the water was within ±20% of the measured mean values.

No occurrence of mortality or abnormality was observed. Surviving fish showed no abnormality in appearance, swimming or feeding behavior. Therefore, test fish were confirmed to be reared and maintained appropriately.

The mean weight of test fish was 5.67 g at the start of the exposure (fish for lipid contents measurement) and 6.38 g at the end of the exposure (all fish taken from the concentration level and control level on day 28).

A steady-state was considered to have been reached at the concentration level, since all the obtained BCF (mean) at each measurement during the exposure period were less than 100 L/kg. A mean BCF of 64 L/kg was determined on Day 28 of the exposure. No depuration-phase was necessary.

The mean BCF value at the end of the 28-day exposure period was 64 L/kg (wet wt.), 63 L/kg (wet wt.) when normalised to 5% lipid content.

All BCF values observed over the 28 day exposure duration were < 100 L/kg (wet wt.), except for one sample on Day 14 which showed a BCF value of 123 L/kg. SCENTAURUS BERRY has a very low potential to bioconcentrate.
