Adequacy of study:
supporting study
Study period:
2008-09-11 to 2009-10-13
Reliability:
2 (reliable with restrictions)
Rationale for reliability incl. deficiencies:
guideline study with acceptable restrictions
Remarks:
GLP study performed according to OECD Guideline 217 (Soil Microorganisms: Carbon Transformation Test) with the following deviation- the test substance characterization was not performed in accordance with GLP. Source: Sodium Tungstate
Target: Ammonium metatungstate
3. CATEGORY APPROACH JUSTIFICATION: See Annex 3 in CSR

AMENDMENT OF SOIL
- Type of organic substrate: Sandy clay loam, collected from Grand Forks North Dakota, April 7, 2009, at a depth of 4-8 inches from a field plot. Stored in a refrigerator at 4 °C until used. Immediately prior to use, soil was passed through a 2mm sieve.
- Other: not applicable

APPLICATION OF TEST SUBSTANCE TO SOIL
- Method: 20 g aliquots (dry weight) of soil were weighed into eighteen 250 mL Pyrex bottles. Bottles were fitted with foam plugs for air exchange. Soil moisture was adjusted to 40-60% of water holding capacity. Soil samples were incubated 6 days at 20 °C. Then, a 20.1 mg/mL stock solution was prepared using 503.9 mg sodium tungstate in a 25 mL volumetric flask, correcting for active ingredient content (99.9%) and bringing to volume with reagent water. Nine samples were dosed with 0.995 mL of the solution. The remaining nine were dosed with the same volume of reagent water only. Each sample was mixed thoroughly.

Results

-Microbial plate counts at Day 0 and Day 28 in the treatment (6.0 x 106, Day 0, 1.2 x 107 Day 28) and control 1.6 x 106 Day 0 and 1.1 x 107 Day 28) indicated that the microbial populations remained active and viable during the course of the study.

-The average respiration rate of the Day 0 test substance was 89.3 mg/kg/hr, which differed from the average control respiration rate of 90.6 mg/kg/hr by -1.2%.

-The average respiration rate of the Day 28 test substance was 89.3 mg/kg/hr, which differed from the average control respiration rate of 92.3 mg/kg/hr by -3%.

-These results indicate that the test substance was not inhibitory to the carbon transformation process.

Validity criteria fulfilled:
yes
Conclusions:
The test substance does not inhibit the carbon tranformation process at concentrations of up to 1000 mg test substance/kg soil.
Executive summary:

No data of sufficient quality on toxicity to soil microorganisms are available for Ammonium metatungstate (target substance). However, data are available for sodium tungstate (source substance), which were used for read-across. Due to similar or lower transformation/dissolution results for the target substance than the source substance, the resulting toxicity potential is expected to be similar or lower, so read-across is appropriate. In addition, read-across is justified because the classification and labelling is the same or less severe for the target substance and the PBT/vPvB profile is the same. Finally, the dose descriptors are, or are expected to be, sufficiently similar or higher for the target substance, and read-across to the source substance is adequately protective. For more details, refer to the read-across category approach description in the Category section of this IUCLID submission or Annex 3 of the CSR.

Description of key information

The toxicity of ammonium metatungstate to soil microorganisms was estimated based on read-across from sodium tungstate. In both a carbon and a nitrogen transformation test using sodium tungstate, the 28-d EC50 was found to be > 1000 mg sodium tungstate/kg soil d. wt. (ca. 586 mg W/kg soil d. wt.) based on nominal concentrations.

Key value for chemical safety assessment

Short-term EC50 for soil microorganisms:
586 mg/kg soil dw
