2-Methylpent-2-enal was assessed for its skin sensitising potential using the Local Lymph Node Assay (LLNA) in mice (BASF SE, 58V0505/11X140). In this study Stimulation Indices (S.I.) of 12.96, 12.89, and 16.37 were determined with the test item at concentrations of 25 and 50% (w/w), and 100% (undiluted test item) in acetone:olive oil (4+1 v/v), respectively. An EC3 value could not be calculated as all S.I.s exceeded the threshold value of 3. The mean DPM value was increased in the high dose group in comparison to the mid and low dose group. A statistically significant and biologically relevant increase in DPM values as well as in lymph node weight and lymph node cell count was observed in all test item treated groups in comparison to the vehicle control group (p<0.05). The cutoff-value for the lymph node cell count index of 1.55 for a positive response was exceeded in all dose groups. The test item 2-Methylpent-2-enal was thus found to be a skin sensitiser under the test conditions of this study.
