STABILITY AND STORAGE CONDITIONS OF TEST MATERIAL

- For determination of the test item concentration, samples were taken from each testing concentration and from the control at the start and at the end of the experiment. Three replicate samples were taken from the test solutions and one control sample on both analytical occasions.

- The test solutions used in the test were prepared by mechanical dispersion without using of any solubilising agent. The test item is poorly water soluble material. In order to achieve maximum dissolved concentration, a supersaturated solution (100 mg/L nominal loading) was prepared by dispersing/dissolving an amount of 0.1 g test item into 1000 mL test medium (OECD medium) approx. one day before the start of the test. This solution was agitated by orbital shaker (~350 rpm) for a period of approx. one day and then the non-dissolved test material was separated by filtration through a fine (0.22 μm) filter in order to obtain the saturated test solution (i.e. 100 % v/v saturated solution). The test solutions of the chosen test concentrations were prepared by appropriate diluting of this stock solution. The test solutions were prepared just before introduction of the algae (start of the experiment).

- Breeding Conditions: The stock cultures are small algal cultures that are planted on agar regularly. These are transferred to fresh medium at least once every two months under standardized conditions according to the test guidelines.

The pre-culture was prepared with Algal Mineral Salts Culture Medium, incubated under the conditions of the test and used when still exponentially growing, normally after an incubation period of about three days. The algal culture flasks were continuously illuminated with fluorescent lamps (with a spectral range of 400-700 nm). The light intensity was checked and recorded at the position occupied by algal culture flasks at the start of the test. The average measured light intensity was 8200 lux. Shape of the algal cells:
No morphological abnormalities were observed in the control and in the two lowest test item concentrations (0.10 and 0.19 mg/L measured) during the experiment. :
Swollen and thin cells were observed amongst the mostly normal algal cells in the three highest concentrations (0.41, 0.91 and 1.65 mg/L measured)
Cell density in control:
In the control the cell density has increased from nominal N= 1 × 104 cells/mL at the start of the test (0 hours) to N= 64.17 × 104 cells/mL (mean value) after 72 hours. Thus, the algal growth in the control was high enough to pass the validity criteria in this assa - Results with reference substance (positive control):
