Test material form:
liquid
Specific details on test material used for the study:
Identification: Vetyveryl Acetate
Decription: Yellow viscous liquid
Batch number: 9000317035
Expiry date: March 25, 2003
Stability of test article: Stable at storage conditions
Storage conditions: In a refrigerator (2 - 8 °C)
Safety precautions: Gloves, goggles and face mask were sufficient to assure personnel health and safety

Test animals / tissue source

Species:
rabbit
Strain:
New Zealand White
Details on test animals or tissues and environmental conditions:
Test system: New Zealand White Rabbit
Rationale: Recognised by the international guidelines as the recommended test system
Source: Elevage Scientifique des Dombes, F-01400 Chatillon sur Chalaronne
Number of animals: 1 male, 2 females
Age at start of treatment: 13 weeks
Identification: By unique cage number and corresponding ear number
Acclimatisation: Under laboratory conditions after health examination. Only animals without any visual signs of illness were used for the study.
Allocation: Male no. 31; Female Nos. 32 - 33

Test system

Vehicle:
unchanged (no vehicle)
Controls:
yes, concurrent no treatment
Amount / concentration applied:
As a liquid, 0.1 ml / animals (per animal) of the test article was measured with a syringe and applied undiluted.
Observation period (in vivo):
Vaibility: Daily
Clinical signs: Daily during the observation period
Body weights: At start of acclimatisation, on the day of application and at termination of observation
Number of animals or in vitro replicates:
3
Details on study design:
The eyes of the animals were examined one day prior to the test article administration.
Animals with overt signs of ocular injury or irritation which may have interfered with the interpretation of the results were not used in the test.
On the day of treatment, the test article (undiluted) was placed in the conjunctival sac of one eye ofeach animal after gently pulling the lower lid away from the eyeball. The lids were then held gently together for about one second to prevent loss of test article. The other eye remained untreated and served as the reference control.

Irritant / corrosive response data:
Application of the test article to healthy rabbit conjunctavae resulted ina primary irritation score of 0.00. In all animals, slight reddening of the conjunctivae as well as moderate watery discharge was observed, one hours after treatment. All signs of irritation were revesible after 24 hours.
Other effects:
No clinical signs of systemic toxicity were observed in the animals during the test and observation period, and no mortality occurred.
No staining by the test article was observe.
No corrosion of the cornea was observed at any of the reading times.
The body weight of all rabbits were considered to be within the normal range of variability.

Applicant's summary and conclusion
