Preparation of stock and test solutions: A stock solution has been prepared with a concentration of 1.0 g/L of the material presented (97.54% active matter). Therefore, 0.1000 g of the material has been dissolved in 100 mL of culture medium in a volumetric flask, Class A of 100 mL by shaking, so we have a stock solution of 975.4 mg/L of active matter . The different solutions used in the test have been prepared from this stock solution of 975.4 mg/L of active material of the test product.

The validation criterion considered for this study is:
- The average number of fronds in the control must present a growth rate specific medium of at least 0.275 per day.
- Duplication time of approximately 2.5 days, and multiplication by 7 of the number of fronds, at the end of the study
- The EC50 7d value of 3,5-diclofenol is in the range 2.2 - 3.8 mg/L.
Results with reference substance (positive control):
The reference substance used to verify the response of the test organisms is 3,5-diclofenol. The result of the periodic verification of the response of the test organisms is EC50 7d = 2.9 mg/L.

Yield inhibitionThe weight of the fronds is determined at the beginning and at the end of the test. The initial weight is calculated from the average of 6 repetitions of weighing 12 fronds. The following table shows the weights obtained for each of the test concentrations.

Conclusions:
The concentration of the test item that produces an inhibition of the 50% growth rate at 7 days (EC50 7d) in Lemna minor is 575 mg/L.
The concentration of the test item that produces an inhibition of Biomass of 50% at 7 days (EC50 7d) in Lemna minor is 103 mg/L.
Executive summary:

The study of acute toxicity to aquatic plants of Direct Black RBB dye was carried out. For the determination of the inhibition of algal growth, the freshwater aquatic plant Lemna minor has been used and the operative method according to the OECD Guideline 221 (2006) has been followed.


The freshwater aquatic plant Lemna minor has been exposed to the substance at the test concentrations 1; 3,2; 10; 32; 100; 320 and 1000 mg/L based on active material. The total duration of the trial has been 7 days.


The result obtained in this test indicates that the dye Direct Black RBB dye presents an inhibition value of the EC50 growth rate (7d) equal to 575 mg/L and an inhibition value of the biomass EC50 (7d) equal to 103 mg/L after 7 days of the test.










 








 





 

Description of key information

There is one study conducted on the substance Direct Black RBB (CAS 68877-33-8) according to the OECD Guideline 221 to determine the toxicity to aquatic plants Lemna minor. Based on the ECHA guidances, this test method is considered more suitable for testing toxicity of coloured test materials to aquatic plants, than the OECD guideline 201 to test the toxicity to aquatic algae.


The EC50 (7d) value 575 mg/L active ingr. obtained in this study, shows that the tested substance is not harmful to this type of aquatic plant.
