PREPARATION AND APPLICATION OF TEST SOLUTION
The test item solutions used in the test were prepared by mechanical dispersion without using of any solubilising agent. A stock solution was first prepared by dissolving an amount of 0.0808 g test item in 808 mL dilution water (OECD Medium, see section 5.4) resulting a nominal concentration of 100 mg/L. Test solutions of subsequent lower concentrations were prepared by appropriate dilution of this stock solution. After the formulation procedure the algal cells were immediately introduced into the test solution (start of experiment).

Reported statistics and error estimates:
Mean values and standard deviations were calculated for each concentration at each observation period using Microsoft Excel for Windows software.
For the determination of the LOEC and NOEC, the calculated mean growth rates and yield at the test concentrations were tested on significant differences to the control values using analysis of variance (ANOVA) and Dunnett-t test (α= 0.05, 2-sided) by SPSS software.
For determination of the ECx values Probit analysis by SPSS software was performed.

Validity of the Test
- The cell density in the control cultures increased by a factor of 149.17 within 72 hours. This corresponds to a specific growth rate of 1.67 day-1.
- The mean coefficient of variation (CV) for section-by-section specific growth rates (days 0-1, 1-2 and 2-3, for 72 h-tests) in the control cultures did not exceed 35 %
    - CV for section-by-section growth rate day 0-1: 8.03 %
    - CV for section-by-section growth rate day 1-2: 15.03 %
    - CV for section-by-section growth rate day 2-3: 9.79 %
The mean coefficient of variation for section-by-section specific growth rates: 10.95 %.
- The coefficient of variation of average specific growth rates during the whole test period (day 0-3) in the control cultures was 1.58 %.

All validity criteria were met, therefore the study can be considered as valid.

Analytical results:

Five different concentrations arranged in a geometric series with a spacing factor of 2 and a concurrent control were tested in the main experiment. The nominal test item concentrations were6.25, 12.5, 25, 50 and 100 mg/L. The measured concentrations of the test item varied between 84 and 99 % of the nominal concentrations (i.e. within ± 20 %) during the experiment, therefore, the biological results are based on the nominal concentrations. In the untreated control group the test item was below the quantitation limit.

Average Specific Growth Rate
The results of the statistical evaluation showed that the 0-72 h average specific growth rate was statistically significantly different from the untreated control value in the concentrations of 25, 50 and 100 mg/L and not significantly different in the concentrations of 6.25 and 12.5 mg/L.
Accordingly, the 72-h NOEC based on growth rate was determined to be 12.5 mg/L and the LOEC was determined to be 25 mg/L.
The 72-h EC10, EC20, EC50 values for growth rate were determined to be 57.1, > 100 and > 100 mg/L, respectively.

Yield
The results of the statistical evaluation showed that the 0-72 h yield was statistically significantly different from the untreated control value in the concentrations of 25, 50 and 100 mg/L and not significantly different in the concentrations of 6.25 and 12.5 mg/L.
Accordingly the 72-h NOEC based on yield was determined to be 12.5 mg/L and the LOEC was determined to be 25 mg/L.

Morphological Changes of Algal Cells
No abnormal shape of algal cells was noticed in the control and in the lowest concentration of 6.25 mg/L during the test.
In the concentrations of 12.5, 25, 50 and 100 mg/L swollen cells were observed together with normal cells during the test.

The algal growth in relation to a control culture was determined over a fixed test period of 72 hours and, thus, over several algal generations. For determination of the test item concentrations, samples were taken from each concentration level and control at the start and at the end of the test. Concentrations were determined using HPLC method with UV detection. The analytically measured test item concentrations remained within ± 20 % of the nominal over the test period of 72 hours, therefore, the biological results are based on the nominal concentrations.
