Details on sampling:
The recovery of the test item in the test medium was assessed at room temperature at nominal concentrations of 1.012 mg/L and 151.000 mg/L, using a validated analytical method. All the test concentrations along with the negative control were analyzed for the test item concentration at the beginning, at 24 hour, at 48 hour and at the end of the test. For the analysis, duplicate set of composite samples were drawn from the prepared test concentrations and from the negative control. The criteria for acceptance of analysis results of test concentration were 80 to 120 % of nominal concentration with ≤20 % RSD of analyzed concentration.

Details on test solutions:
Test item stock of 1 mg/mL was prepared by grinding 500 mg test item in test medium and made up to 500 mL in a volumetric flask using test medium and sonicated for 30 minutes. Control (G1) was prepared by adding 3.2 mL algal suspension to achieve initial cell density of 1x10E4 cells/mL and made up to 1 L in a volumetric flask using test medium. To prepare test concentrations of 3.125 (G2), 6.25 (G3), 12.5 (G4), 25 (G5) and 50 (G6) mg/L, 3.125, 6.25, 12.5, 25 and 50 mL of test item stock was mixed with test medium, 3.2 mL algal suspension was added to achieve initial cell density of 1x10E4 cells/mL and made up to 1 L in a volumetric flask using test medium separately. 150 mL of prepared negative control was dispensed into 6 test containers and 150 mL of each test item concentration was dispensed separately into 3 test containers.

Culture Maintenance:
Algal cells were maintained at the test facility through sub-culturing once in 5 to 10 days from the continuous culture into fresh medium in a conical flask. These conical flasks were incubated between 21 and 25 °C (± 2 °C) under a white fluorescent continuous illumination of 8000 ± 1600 Lux.

Test Item Solubility


FAT 75637/B was tested for its solubility in the test medium and formed semi-transparent cream coloured formulation with visible solid particles in test medium at 50 mg/L.


Definitive test result:


At the end of the test (72 hours), the cell biomass in the test item solutions decreased with the increase in test concentration. At test concentration of 3.125, 6.25, 12.5, 25 and 50 mg/L, the observed inhibition in the growth rate of algal biomass was 0, 0, 20.06, 21.54 and 24.82 %; and the percent yield inhibition was 0, 0, 59.18, 61.82 and 67.11 %, respectively when compared to that of the negative control at the end of the 72 hour test period. Observed cells were found morphologically normal in all the tested concentrations at the end of test. The EC10 and EC20 values for growth were between 6.25 mg/L (5.94 mg a.i./L) and 12.5 mg/L (11.88 mg a.i./L) based on the nominal concentrations. The EC50 value for growth rate was higher than 50 mg test item/L based on the nominal concentrations. The EC10, EC20 and EC50 values for yield were between 6.25 mg/L (5.94 mg a.i./L) and 12.5 mg/L (11.88 mg a.i./L) based on the nominal concentrations.


VALIDITY OF TEST


There was an increase in cell concentration of the negative control culture by a factor of 78.79, which is more than the required factor limit of at least 16 at the end of the test. The mean coefficient of variation for section-by-section specific growth rates in the negative control cultures during the test was 27.64 %, which is within the required limit of 35 %. The coefficient of variation of average growth rate between replicate cultures of negative control was 0.27 , which is within the required limit of 7 %.
