- Sampling method: All loading rates and the control were analytically verified via LCMS at the start (0 hours) of the exposure (72 hours) with algae.
At the start, samples were taken from one additional replicate of each test item concentration and the control and at the end samples were taken from pooled replicates.

PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
- Method:
Preparation of the stock solutions
A saturated solution with a nominal loading rate of 100 mg test item/L was freshly prepared and treated with ultrasound for 5 minutes at room temperature. Thereafter the solution was stirred for 4 hours (room temperature, 1100 rpm) with a magnetic stir rer. Undissolved particles were removed by membrane filtration The filter was saturated in order to avoid adsorption during the filtration. The first 25 mL of the filtrate were discarded. The filtration was interrupted for 15 minutes to allow adsorption and saturation of the filter material with dissolved test item. Thereafter, the filtration was continued. The next 25 mL were discarded. The following filtrate, i.e. the saturated solution, was used in the test. During filtration, the filter was always kept covered. The Tyndall effect was positive.

Preparation of the test item concentrations
Out of the saturated solution five dilution levels were tested in a geometrical series with a dilution factor of √10: 1.00 - 3.16 - 10.0 - 31.6 - 100% of the saturated solution, corresponding to the geometric mean measured test item concentrations of: 0.686 –
2.19 – 6.66 – 20.7 – 65.7 mg test item/L. The dilution levels are based on the results of a preliminary range finding test (non-GLP).

- Controls: Six replicates (without test item) were tested under the same test conditions as the test item replicates.

The cells were checked for unusual cell shapes, colour differences, differences in chloroplast morphology, flocculation, adherence of algae to test containers and agglutination of algae cells.

The NOEC / LOEC was determined by calculation of statistical significant differences of growth rate and yield using Dunnett’s Method. A Shapiro-Wilk’s Normality test and a Brown-Forsythe’s Equal Variance test were done first. P-values for both Normality
and Equal Variance tests are 0.05. The α -value (acceptable probability of incorrectly concluding that there is a difference) is α = 0.01.

In this study, the effect values to the test item on the growth of the freshwater green alga Pseudokirchneriella subcapitata after 72 h were determined as follows (based on geometric mean measured test item concentrations): The EC50-values for inhibition of growth rate (ErC50) and yield (EyC50) after 72 h were 24.4 mg/L and 6.68 mg/L, respectively. The NOEC for both growth rate and yield was 2.19 mg/L, respectively.
