Materials and methods

Test guideline
Qualifier:
no guideline followed
Principles of method if other than guideline:
Female albino rats received intracranial injections of 2,2'-bipyridine into the substantia nigra or into the nucleus caudatoputamen. In parallel, mixtures of 2,2'-bipyridine with ammonium iron(II) sulfate were tested. Tremor resulting from these injections was assessed after 0.5, 1, 3, and 5 h after injection using a 4-stage rating system. Dopamine content of the striatum was also quantified.

Results and discussion

Any other information on results incl. tables

Female rats, when injected 2,2'-bipyridine (alone or in combination with ammonium iron(II) sulfate) into the substantia nigra showed slight but significant tremor compared to control animals (Table 1). No effect occurred when the test item was injected into the nucleus caudatoputamen. The dopamine content of the striatum remained unchanged upon injection (Table 2).

Applicant's summary and conclusion

Conclusions:
When rats are given 2,2'-bipyridine (alone or in combination with ammonium iron(II) sulfate) as injection into the substantia nigra, tremor is induced. Influence on striatal dopamine content has not been found.
Executive summary:

Female albino rats received intracranial injections of 2,2'-bipyridine into the substantia nigra or into the nucleus caudatoputamen. In parallel, mixtures of 2,2'-bipyridine with ammonium iron(II) sulfate were tested. Tremor resulting from these injections was assessed using a 4-stage rating system. Dopamine content of the striatum was also quantified.

Injection of 2,2'-bipyridine (alone or in combination with ammonium iron(II) sulfate) into the substantia nigra showed slight but significant tremor compared to control animals; no effect was seen after injection into the nucleus caudatoputamen. The dopamine content of the striatum remained unchanged upon treatment.
