The aquatic toxicity of the 'ethylene glycol and higher glycols' (mono-, di-, tri-, tetra- and pentaethylene glycol) is evaluated in a read-across approach.

The glycol read-across includes the assessment for the described reaction mass of 2,2'-(ethylenedioxy)diethanol 3,6,9-trioxaundecane-1,11-diol, as a glycole mixture. Further information on the read-across approach is described in a separate document, which is attached to this IUCLID-File in Chapter 13.

Data on all three trophic levels (fish, daphnia, algae) are available to describe the aquatic toxicity of the glycol read-across members. The available data cover short and long-term effects on all three trophic levels. Due to the fact, that not for each single substance data for all required endpoints are available, a weight of evidence approach is used, which includes additional information based on QSAR calculation with the EpiWin-Program ECOSAR v1.11. Measured data as well as estimated data demonstrate, that all glycols within the read-across are not harmful to aquatic organisms. No adverse effects on aquatic organisms occurred up to concentrations above 100 mg/L. 

From the QSAR calculations it can be expected that algae are slightly more sensitive (ChV = 479 - 8450 mg/L) than invertebrates (ChV = 690 – 17475 mg/L) or fishes (ChV = 2629 – 8666 mg/L). Therefore, the chemical safety assessment is based on the algae data. Within the read-across several studies on algae are available. In one case (pentaethylene glycol, CAS 4792-15-8) only a limit test with a concentration of 100 mg/L was conducted, in which no effects were observed. From the other available information it can be expected that the real NOEC is far above the limit test concentration of 100 mg/L. Nevertheless, as a worst-case assumption the limit test concentration of 100 mg/L was used as NOEC value for the PNEC derivation.
