Sampling and analysis

Analytical monitoring:
yes
Details on sampling:
- Concentrations: one limit concentration 100 mg/L plus the control
- Sample storage conditions before analysis: The sample of the test item was stored at ambient temperature without exposure to light in a tightly
sealed container.

Test solutions

Vehicle:
no
Details on test solutions:
PREPARATION AND APPLICATION OF TEST SOLUTION (especially for difficult test substances)
The test item in the amount of 100.70 mg was weighed in a glass crystallizer transferred by multiple washing with Elendt M7 medium into a glass flask, the total volume was filled up to 1007 mL. The test item concentration of 100 mg/L was visually homogeneous
and transparent. After 24 h of exposure the test item concentration and the control were renewed to
freshly prepared.

Test organisms

Test organisms (species):
Daphnia magna
Details on test organisms:
TEST ORGANISM
- Common name: Daphnia magna Straus
- Source: Institute of Industrial Organic Chemistry, Branch Pszczyna, Department of Ecotoxicology
- Age at study initiation (mean and range, SD): 24 hours old (not first brood progeny)
- Method of breeding: Daphnia magna was cultured in glass beakers with a capacity of 150 mL (one parent per vessel)
containing 100 mL of Elend M7 medium, at room temperature ranging from 18 to 22°C with daily cycle
16 h light : 8 h dark
- Feeding during test: no feeding

ACCLIMATION

- Type and amount of food: suspension of algae; mixture of two species Pseudokirchneriella subcapitata :
Desmodesmus subspicatus (in 2:1 ratio) Group B vitamins and micronutrients necessary for proper growth
were supplied with the lyophilized suspension of Spirulina sp.

20.4 - 21.9 ºC
pH:
7.31 - 7.46
Dissolved oxygen:
8.65 - 9.01 mg/L
Nominal and measured concentrations:
nominal test item concentrations: 100 mg/L plus the control
Details on test conditions:
TEST SYSTEM
- Test vessel: glass beakers of 150 mL with transparent lids
- Type (delete if not applicable): closed
- Aeration: no aeration.
- Renewal rate of test solution (frequency/flow rate): one renewal
- No. of organisms per vessel: five Daphnia magna in each replicate
- No. of vessels per concentration (replicates): 8
- No. of vessels per control (replicates): 8

TEST MEDIUM / WATER PARAMETERS
- The Elendt M7 medium recommended by the OECD Guideline No. 202 (2004) was used to culture the
test organism and as a diluent/solvent of the test item

OTHER TEST CONDITIONS

- Light intensity:daily cycle 16 h light : 8 h dark; fluorescent light source

EFFECT PARAMETERS MEASURED (with observation intervals if applicable) :
Daphnia magna was observed for immobilization after 24 and 48 h of exposure. The Daphnia magna
was considered immobile if they showed no ability to swim within 15 seconds after swirling the test
vessel

TEST CONCENTRATIONS

- Test concentrations: were determined on the basis of the preliminary tests results
Reference substance (positive control):
yes
Remarks:
potassium dichromate

Applicant's summary and conclusion

Validity criteria fulfilled:
yes
Conclusions:
In the definitive test the validity criteria were met according to OECD Guideline No. 202 (2004):
- The percentage of immobilization of Daphnia magna in the control was 5% (criterion: not more
than 10%),
- The dissolved oxygen concentrations in the test vessels were within the range of
8.62 – 9.01 mg/L (criterion: not less than 3 mg/L)
