Study meets the critria for Klimisch score 2 (reliable with restrictions) because it followed an internationally recognised method (OECD Guideline 305) and was carried out to Good Laboratory Practice. However, this study was only available for review as a robust study summary.
Justification for type of information:
The sulfonate group is a common functional group present in each of the analogue members, and is expected to exhibit similar biological activities with little influence from the length of carbon chain. Any potential breakdown products, via physical or biological processes, are also expected to result in structurally similar chemicals. In addition, non-random patterns have been observed for the toxicological effects (e.g., available data showed low levels of acute toxicity, lack of mutagenic potential, skin irritating properties and a trend of change in ecotoxicological potential based on molecular weight). These common behaviors and consistent trends suggest a common mechanism and mode of action thereby providing further supporting evidence for the read-across among the linear and branched alkyl benzene sulfonates.

Details on test conditions:
Rainbow trout were exposed in groups of 52 to an aqueous solution of test material for 42 days under dynamic conditions. Samples of test fish were taken from the solvent control, 0,001 and 0.01 mg/l test groups on days 21, 25, 28, 32, 35 and 39 of the uptake phase, and concentrations of the test material in fish tissues were determined. After 42 days exposure, the remaining fish were subjected to a depuration period of 61 days. Samples of test fish were taken from the solvent control, 0.001 and 0.01 mg/l test groups on days 3, 6, 11, 13 and 61 of the depuration phase and the concentrations of the test material determined.

14-C SP6012 (oil free) was not significantly accumulated in fish tissue and was observed to be eliminated over a 61-day depuration period. At the end of the depuration period, 70 and 90% of 14C-SP6012 (oil free) was eliminated from the fish tissues at concentrations of 0.001 and 0.01 mg/l, respectively. Refer to Tables 1 and 2

Conclusions:
14-C SP6012 (oil free) was not significantly accumulated in fish tissue and was observed to be eliminated over a 61-day depuration period. At the end of the depuration period, 70 and 90% of 14C-SP6012 (oil free) was eliminated from the fish tissues at concentrations of 0.001 and 0.01 mg/l, respectively.

In a biconcentration study, rainbow trout (Oncorhynchus mykiss) were exposed to 14-C SP6012 (oil free) at concentrations of 0.001 and 0.01 mg/L under dynamic conditions conditions. 14-C SP6012 (oil free) was not significantly accumulated in fish tissue and was observed to be eliminated over a 61-day depuration period.

 

This toxicity study is classified as acceptable and satisfies the guideline requirement for a fish bioaccumulation study.

In a bioconcentration study performed according OECD Guideline 305 (Bioconcentration: Flow-through Fish Test), rainbow trout (Oncorhynchus mykiss) were exposed to 14-C SP6012 (oil free) at concentrations of 0.001 and 0.01 mg/L under dynamic conditions. 14-C SP6012 (oil free) was not significantly accumulated in fish tissue and was observed to be eliminated over a 61-day depuration period. The calculated BCF was 45 and 38 at 0.001 mg/l (steady state and kinetic state respectively) and 58 and 64 at 0.01 mg/l (steady state and kinetic state respectively).

This toxicity study is classified as acceptable and satisfies the guideline requirement for a fish bioaccumulation study.

The prediction of the BCF performed with the computer program BCFBAF v3.01 (EPIWIN software) by US-EPA
